A phylogenetic survey of myotubularin genes of eukaryotes: distribution, protein structure, evolution, and gene expression [PDF]
Background Phosphorylated phosphatidylinositol (PtdIns) lipids, produced and modified by PtdIns kinases and phosphatases, are critical to the regulation of diverse cellular functions.

Myotubularin Related Protein 7, a Novel Stim1 Binding Protein
Stromal interaction molecule 1 (STIM1) is a Ca2+ sensor in the endoplasmic reticulum (ER) membrane. The protein plays a crucial role in store-operated Ca2+ entry (SOCE) by transducing ER Ca2+ depletion signals to Ca2+ release-activated Ca2+ channel protein 1 (ORAI1) at the plasma membrane.

Deficiency of myotubularin-related protein 14 influences body weight, metabolism, and inflammation in an age-dependent manner [PDF]
Myotubularin-related protein 14 (MTMR14) is a novel phosphoinositide phosphatase with roles in the maintenance of normal muscle performance, autophagy, and aging in mice. Our initial pilot study demonstrated that MTMR14 knock out (KO) mice gain weight earlier than their wild-type (WT) littermates, which suggests that this gene may also be involved in ...

Myotubularin-related protein 1 (MTMR1) is a phosphatase that belongs to the tyrosine/dual-specificity phosphatase superfamily. MTMR1 has been shown to use phosphatidylinositol 3-monophosphate (PI(3)P) and/or phosphatidylinositol 3,5-bisphosphate (PI(3,5)P2) as substrates. Here, we determined the crystal structure of human MTMR1.

ABSTRACT Background Autosomal dominant centronuclear myopathy (ADCNM), most commonly caused by mutations in the dynamin 2 (DNM2) gene, is a rare congenital myopathy characterized by progressive muscle weakness and atrophy. Myostatin, a key negative regulator of skeletal muscle mass, has shown therapeutic potential in several models of neuromuscular ...

Protein-protein interactions between myotubularin and MTMR12 proteins.
(A) GST pull down of MTM1-GST recombinant protein with MTMR12 showed a direct interaction between the two proteins (above). Coomassie blue stained gel (below) showing GST and MTM1-GST protein used in the pull down (B) Co-IP experiments from Cos-1 ...

A 24‐year‐old heterozygous woman presented with lifelong hypotonia and slowly progressive, asymmetric axial and limb‐girdle weakness with facial/ocular involvement and restrictive ventilatory impairment (FVC 53.12% predicted). Multimodal evaluation (EMG, muscle MRI, and whole‐exome sequencing [WES]) identified a de novo MTM1 frameshift variant ...

Targeting PIKfyve: Chemical Biology and Drug Discovery Insights
PIKfyve inhibitors have progressed from niche chemical probes to powerful tools for interrogating endolysosomal biology, enabled by rapid and robust cellular phenotypes. Compounds such as apilimod establish PIKfyve as a druggable lipid kinase, while agents like vacuolin‐1 underscore the importance of distinguishing true target engagement from ...
