New approach methodologies for evaluating developmental and reproductive toxicity potential: Review on state of the art, applicability for cosmetic related ingredients and coverage of developmental stages

open access: yesNAM Journal
The development of in vitro developmental and reproductive toxicity (DART) models is challenging. We made a brief review of in silico models and an in-depth review of 17 established in vitro New Approach Methodologies (NAMs) used to assess the DART ...

Wave propagation in a nonlinear acoustic metamaterial beam considering third harmonic generation

open access: yesNew Journal of Physics, 2018
Nonlinear acoustic metamaterials (NAMs) provide new ways to control elastic waves. In this work, flexural wave propagation in an infinite NAM beam consisting of periodic Duffing resonators is reported by considering the third harmonic generation ...

Empirical PK/PD Model for Differentiating Intrinsic Dosing Rules by Therapeutic Indication With Relevance for HIV‐1 Latency Reversal

open access: yesClinical Pharmacology &Therapeutics, Volume 120, Issue 5, Page 1414-1426, November 2026.
Romidepsin (RMD), an FDA‐approved histone deacetylase inhibitor for treating cancer, demonstrates potent latency‐reversing activity in vitro but has yielded suboptimal outcomes in kick‐and‐kill HIV‐cure trials when administered as a 5 mg/m2 infusion.

Lung microphysiological system for predicting antiviral drug efficacy for COVID-19

open access: yesThe Microbe
Development of physiologically relevant in vitro models that recapitulate key aspects of SARS-CoV-2 infections has proven challenging due to limitations associated with current New Approach Methodologies (NAMS).

Target‐Engagement Prediction From Molecular Structure, In Vitro and Animal Data: A Tutorial for Early Assessment of Clinical Drug Exposure and Therapeutic/Adverse Potential Using the ANDROMEDA and PLATO Software on the InSilicoTrials Platform

open access: yesCPT: Pharmacometrics &Systems Pharmacology, Volume 15, Issue 11, November 2026.
ABSTRACT The target‐engagement predictor, implemented within the InSilicoTrials cloud‐based, AI‐powered platform for model‐informed drug discovery and development, enables early exploration of time‐dependent pharmacological responses. Predictions can be generated using inputs ranging from molecular structure to in vitro, animal, and clinical data ...

New approach methods to assess developmental and adult neurotoxicity for regulatory use: a PARC work package 5 project

open access: yesFrontiers in Toxicology
In the European regulatory context, rodent in vivo studies are the predominant source of neurotoxicity information. Although they form a cornerstone of neurotoxicological assessments, they are costly and the topic of ethical debate.

Potentiation effects of the positive allosteric modulator PTC‐174 on NMDA receptors in neonatal male rat substantia nigra dopaminergic neurons

open access: yesBritish Journal of Pharmacology, Volume 183, Issue 21, Page 6481-6500, November 2026.
Abstract Background and Purpose Positive allosteric modulators bind to secondary binding sites on the receptor and alter receptor conformation and activation, changing agonist potency and efficacy. PTC‐174 is a novel NMDA receptor positive allosteric modulator that particularly enhances activity of receptors containing GluN2C or GluN2D subunits.

In Situ Observation of Pressure‐Enhanced Hydrous–Anhydrous Solid Solution in Lower‐Mantle SiO2–AlOOH Phases

open access: yesGeophysical Research Letters, Volume 53, Issue 19, 16 October 2026.
Abstract Water plays a critical role in Earth's mantle dynamics and evolution, yet lower‐mantle storage mechanisms remain debated. δ‐AlOOH and SiO2 phases have been independently proposed as high‐pressure water hosts. Here, we conducted in situ synchrotron X‐ray diffraction experiments with a multi‐anvil apparatus and sintered diamond anvils to ...

Evolutionäre, funktionelle und mechanistische Charakterisierung mikrobieller Terpensynthase‐ähnlicher Gene aus Springschwänzen

open access: yesAngewandte Chemie, Volume 138, Issue 41, 5 October 2026.
