Nectin-4 Expression Patterns and Therapeutics in Oncology

Cancer Letters
Nectin-4 is a transmembrane receptor that is implicated in migration, adhesion, and proliferation of tumor cells. It has minimal expression in healthy adult tissue but is overexpressed in a number of cancers, with higher expression levels seen in breast, bladder, colorectal, gallbladder, gastric, and non-small cell lung cancers.

Nectin‐4 Expression in Prostatic Adenocarcinoma: An Immunohistochemical Study

The Prostate
ABSTRACTBackgroundThe Nectin‐4 directed antibody drug conjugate enfortumab vedotin (EV) has emerged as frontline systemic therapy in combination with immune checkpoint blockade for urothelial carcinoma (UC), capitalizing on the ubiquitous expression of this protein in UC.

Exploring Nectin-4 expression in hepatocellular carcinoma.

Journal of Clinical Oncology
632 Background: Nectin cell adhesion protein 4 (Nectin-4) is overexpressed in various malignancies, contributing to cancer progression and poor prognosis. We aimed to assess Nectin-4 expression in hepatocellular carcinoma (HCC).

The biology and rationale of targeting nectin-4 in urothelial carcinoma

Nature Reviews Urology, 2020
Bladder cancer is the tenth most common cancer type worldwide. Urothelial carcinoma is the most common type of bladder cancer and accounts for 90% of bladder cancer cases in the USA and Europe. Novel approaches are needed to improve patient outcomes. Nectin-4 is a tumour-associated antigen found on the surface of most urothelial carcinoma cells. In the

Nectin-4 Positivity in Genitourinary Malignancies: A Systematic Review

JCO Precision Oncology
PURPOSE Aberrant expression of nectin-4 (N4) has been observed in several malignancies emerging as new target for antibody-drug conjugates, especially in urothelial carcinoma of the bladder (UBC). Limited data on N4 positivity in nonurothelial genitourinary (GU) cancers are available. This systematic-review aimed to investigate

Abstract 6029: 225Ac/89Zr labeled anti nectin 4 radioimmunoconjugates as theranostics against nectin 4 positive triple negative breast cancer

Cancer Research
Abstract Nectin4 (N4) is a biomarker that is overexpressed in ~62% of triple negative breast cancer (TNBC) and its lack of expression in normal tissues makes it an ideal target for the therapy and PET imaging of TNBC. Methods: We developed a fully human antibody against nectin 4 (anti N4) using phage display.

The diagnostic efficacy of nectin 4 expression in ovarian cancer patients

Biomarkers, 2014
Nectin 4 plays a significant role in cancer cell growth and invasion. To explore its diagnostic role in ovarian cancer.Evaluation of nectin 4 expression in ovarian biopsies by real-time PCR and serum nectin 4 and CA-125 by ELISA in 39 ovarian cancer patients, 21 females with benign ovarian neoplasms and 25 control females.Nectin 4 expression was ...

Expression of nectin-4 in bladder cancer with variant histology.

Journal of Clinical Oncology, 2020
546 Background: The antibody-drug conjugate enfortumab vedotin is poised to change the bladder cancer (BC) treatment landscape by targeting Nectin-4, near ubiquitously expressed in urothelial cancer (UC). Less is known about this and other targets in BC with pure or mixed variant histology (VH).

A chicken homologue of nectin-4 functions as a measles virus receptor

Vaccine, 2016
Measles virus (MV) vaccine strains use CD46, signaling lymphocyte activation molecule, and nectin-4 in human cells as receptors. Meanwhile, many of them are propagated in primary chicken embryonic fibroblasts (CEFs). Our data revealed that CEFs express a nectin-4 homologous molecule (CEF nectin-4) containing well-conserved motifs in the FG and BC loops,

Molecular Crosstalk Between Adherens Junction Proteins, E-cadherin and Nectin-4

Journal of Molecular Biology
Cell-cell junctions formed by the association of cell adhesion molecules facilitate physiological events necessary for growth and development of multicellular organisms. Among them, cadherins and nectins organize and assemble to form adherens junction, which thereby mechanically couples interacting cells.
