Loss of SETD2 in wild‐type VHL clear cell renal cell carcinoma sensitizes cells to STF‐62247 and leads to DNA damage, cell cycle arrest, and cell death characteristic of pyroptosis

open access: yesMolecular Oncology, EarlyView.
Vulnerability to the small molecule STF‐62247 is observed in SETD2‐inactivated VHL‐positive clear cell renal cell carcinoma (ccRCC) cells. DNA damage and loss of CDC25A leads to cell cycle arrest in S phase. Cleavage of caspase‐3 (CASP3) and gasdermin E (GSDME) suggests cell death by pyroptosis.

The CDK12–BRCA1 signaling axis mediates dinaciclib‐associated radiosensitivity through p53‐mediated cellular senescence

open access: yesMolecular Oncology, EarlyView.
Dinaciclib blocks CDK12 activity, leading to downregulation of BRCA1, and probably other proteins involved in homologous recombination, promoting nonhomologous end joining. In this scenario, the presence of ionising radiation triggers a p53‐dependent senescent response that yields increased radiosensitivity. Pan‐cyclin‐dependent‐kinase (CDK) inhibitors

‘soundAFFECTs’: translation, writing, new media, affect

open access: yes, 2009
This chapter will focus on the translation or trans-coding of a work from its print form as ‘AFFECTions: friendship, community, bodies’ (Brewster and Smith 2003) into its multimedia form as ‘soundAFFECTs’ (Dean et al. 2004). In particular, it will explore how this transition changes the affective experience the piece transmits.

The PTTG1/VASP axis promotes oral squamous cell carcinoma metastasis by modulating focal adhesion and actin filaments

open access: yesMolecular Oncology, EarlyView.
VASP was found to be overexpressed in metastatic oral squamous cell carcinoma (OSCC) tissues. Notably, PTTG1‐ and VASP‐deficient OSCC cells demonstrated suppressed metastatic properties by disrupting the interaction between the cytoskeleton and focal adhesion (FAs) in the filopodia region.

Obesity alters the fitness of peritumoral adipose tissue, exacerbating tumor invasiveness in renal cancer through the induction of ADAM12 and CYP1B1

open access: yesMolecular Oncology, EarlyView.
Tumor microenvironment drives cancer formation and progression. We analyzed the role of human cancer‐associated adipocytes from patients with renal cell carcinoma (RCC) stratified as lean, overweight, or obese. RNA‐seq demonstrated that, among the most altered genes involved in the tumor–stroma crosstalk, are ADAM12 and CYP1B1, which were proven to be ...
