Nitroreductases in Prodrug Therapies

2020
The aim of prodrug design is depended on optimizing the conditions to generate rational drug. The nitroreductases have been developed for prodrug strategies and nitro aromatic compounds are mostly used in the enzyme/prodrug combinations. Nitroreductase genes found most commonly in bacterial genomes and nitroreductase-like proteins also found rarely in ...

Activity of an NADPH-Dependent Nitroreductase in Houseflies

Science, 1971
A nitroreductase, reducing parathion to aminoparathion, was found in the soluble fraction that was obtained from abdomens of female houseflies. The reaction required reduced nicotinamide adenine dinucleotide phosphate (NADPH), but was not affected by the presence or absence of oxygen.

Nitroreductase-based GDEPT

Current Pharmaceutical Design, 2002
Nitroreductases that metabolise aromatic nitro groups to hydroxylamines are attractive as enzymes for GDEPT because of the very large electronic change that this metabolism generates, providing an efficient switch that can be exploited to generate potent cytotoxins. While nitroreductase enzymes are widespread, nearly all the work using these in GDEPT

Identification of Aldehyde Oxidase as the Neonicotinoid Nitroreductase

Chemical Research in Toxicology, 2005
Imidacloprid (IMI), the prototypical neonicotinoid insecticide, is used worldwide for crop protection and flea control on pets. It is both oxidatively metabolized by cytochrome P450 enzymes and reduced at the nitroguanidine moiety by a previously unidentified cytosolic "neonicotinoid nitroreductase", the subject of this investigation.

Nitroreductase-activated nitric oxide (NO) prodrugs

Bioorganic & Medicinal Chemistry Letters, 2013
Due to the involvement of nitric oxide (NO) in numerous and diverse physiological processes, site-directed delivery of therapeutic NO in order to minimize unwanted side-effects is necessary. O(2)-(4-Nitrobenzyl) diazeniumdiolates are designed as substrates for Escherichia coli nitroreductase (NTR), an enzyme that is frequently used to facilitate ...

Retro-Nitroreductase, a Putative Evolutionary Precursor to Enterobacter cloacae Strain 96-3 Nitroreductase

Antioxidants & Redox Signaling, 2001
Enterobacter cloacae strain 96-3 nitroreductase (NR) is a homodimeric flavoenzyme that catalyzes the pyridine nucleotide-dependent four-electron reduction of a variety of nitroaromatic compounds, including the explosives TNT (2,4,6-trinitrotoluene), RDX (1,3,5-trinitro-1,3,5-triazine), tetryl (2,4,6-trinitrophenyl-N-methylnitramine), and pentryl (2,4,6-
