Compact 9dBEs Enable Efficient and Precise Genome Editing in Mammalian Cells and In Vivo
As a compact type II‐D system, the Cas9d‐based platform holds great potential for in vivo applications. Through rational engineering, its derived base editors (9dBEs) enable efficient disease modeling while facilitating single‐vector AAV delivery for in vivo genome editing. These miniature tools offer a robust strategy for basic research and biomedical

Streptococcal mannose phosphotransferase system component IID (Man‐PTSIID) is identified as a novel RANK‐binding osteoclastogenic factor. By directly binding to RANK and activating NF‐κB independently of TLR2, Man‐PTSIID drives osteoclastogenesis and inflammatory bone destruction, uncovering an unexpected microbial mechanism underlying streptococcal ...

Switching Spike Plasticity Shapes ACE2 Engagement Across SARS‐CoV‐2 Variants
Conformational plasticity governs SARS‐CoV‐2 spike function and ACE2 recognition. Using high‐speed AFM and single‐molecule force spectroscopy, we analyzed the ancestral spike and nine variants, revealing progressive rigidification from Delta, variable plasticity in Omicron sublineages, and JN.1 compaction.

Chromosome 16q loss drives genomic instability through disruption of the CYLD–TIRR–53BP1 axis. CYLD preserves homologous recombination by stabilizing TIRR and limiting 53BP1 accumulation at DNA double‐strand breaks. CYLD deficiency redirects repair toward error‐prone non‐homologous end joining, promotes mutational burden and homologous recombination ...

Initiation of Protein Synthesis with Non-Canonical Amino Acids In Vivo
AbstractBy transplanting identity elements into E. coli tRNAfMet, we have engineered an orthogonal initiator tRNA (itRNATy2) that is a substrate for Methanocaldococcus jannaschii TyrRS. We demonstrate that itRNATy2 can initiate translation in vivo with aromatic non‐canonical amino acids (ncAAs) bearing diverse sidechains.

Incorporation of non-canonical amino acids into proteins in yeast
Fungal Genetics and Biology, 2016Non-canonical amino acids add extraordinary chemistries to proteins when they gain access to translation. In yeast, they can be incorporated into proteins by replacing a canonical amino acid or in a site-specific manner in response to an amber stop codon.

Incorporation of Non-Canonical Amino Acids into Proteins by Global Reassignment of Sense Codons
Non-canonical amino acids are finding increasing use in basic and applied research. Proteins that evolved naturally for biological function did so by exploiting the chemistries of the canonical amino acids; however, when proteins are repurposed for ...
