Design and Synthesis of Polyamine‐Proteolysis Targeting Chimera Conjugates for Histone Deacetylase (HDAC) Degradation with Enhanced Cellular Uptake

In this article, proteolysis targeting chimeras (PROTACs) bearing a polyamine linker capable of crossing cancer cell membranes via the polyamine transport system are synthesized and evaluated. The results provide valuable insights for the design of PROTACs with enhanced cellular uptake efficiency.

This study investigated airway epithelial barrier dysfunction persistence in ex‐smokers with COPD and underlying mechanisms using single‐cell RNA and ATAC sequencing. Epigenetic NKX2‐1/SPDEF dysregulation maintains goblet cell hyperplasia and barrier dysfunction despite long‐term smoking cessation.

Nanobody–chlorin e6 conjugate for Nectin‐4‐mediated tumor targeting and enhanced photodynamic therapy

Nectin‐4 is frequently overexpressed in solid tumors, yet many PDT photosensitizers show poor water solubility and nonspecific uptake. A Nectin‐4‐targeted nanobody–chlorin e6 conjugate (NPC) improves aqueous solubility and selectively accumulates in Nectin‐4‐positive tumor cells.
