腺病毒介导神经营养素-3 基因转染施万细胞

【目的】构建神经营养素-3(NT-3)基因转染的施万细胞。【方法】NT-3基因重组腺病毒(AdvNT-3)扩增、纯化和测定后,转染体外培养的施万细胞(SCs)。用免疫细胞化学和ELISA方法分别检测NT-3基因转染SCs的NT-3表达及培养液中NT-3的含量。【结果】实验中获得的AdvNT-3中外源基因序列与大鼠NT-3的DNA序列完全一致,与未基因转染SCs相比,NT-3基因转染SCs的NT-3阳性增强,培养液中NT-3含量增高。【结论】通过腺病毒介导可以获得NT-3过量表达的基因转染SCs。

This doctoral thesis shows for the first time that neurotrophin-3 (NT3) and its main receptor, TrkC, are expressed in human visceral adipose tissue (AT), and in white and brown adipose tissue (WAT and BAT respectively) of rodents. NT3 expression is lower in adipocytes than in the vessels that irrigate the tissue.
