Ribonucleotide Scanning Quantification sequencing (RiSQ‐seq) enables absolute quantification of misincorporated ribonucleotides (rNMPs) in the genome by competitive sequencing between rNMPs and backgrounds. The rNMP quantification revealed non‐uniform incorporation and repair activity of rNMP.
