A novel self‐assembled nanomedicine (PMDDH) has been developed for the co‐delivery of doxorubicin and metformin, aiming to enhance the anti‐tumor efficacy of doxorubicin while alleviating its cardiotoxicity through the activation of cGAS‐STING and AMPK signaling pathways, respectively.

Targeted DNA Nanomachine Enables Specific miRNA‐Responsive Singlet Oxygen Amplification for Precise Cutaneous Squamous Cancer Therapy

An intelligent DNA nanomachine based on DNA tetrahedron is proposed, incorporated with aptamer and specific miRNA‐responsive hairpin catalytic assembly (HCA), for precise therapy of cutaneous squamous cell carcinoma (cSCC) in mice, with barely systemic toxicity and inflammation.

MeCP2 Lactylation Protects against Ischemic Brain Injury by Transcriptionally Regulating Neuronal Apoptosis

MeCP2 lactylation protects against ischemic stroke by reducing brain infarct volume and improving neurological outcomes. Lactylation at K210/K249 exerts neuroprotective effects by repressing the transcription of apoptosis‐associated genes, including Pdcd4 and Pla2g6. HDAC3 and p300 serve as key regulatory enzymes for MeCP2 lactylation following stroke.

Diallyl Trisulfide From Garlic Regulates RAB18 Phase Separation to Inhibit Lipophagy and Induce Cuproptosis in Hepatic Stellate Cells for Antifibrotic Effects

Diallyl trisulfides (DATs) selectively induce cuproptosis in hepatic stellate cells (HSCs) by targeting Ras‐related protein Rab‐18 (RAB18) and regulating lipophagy. DATs promote RAB18 phase separation, enhance mitochondrial‐associated membrane structures (MAMs) formation, and increase succinylation of dihydrolipoamide dehydrogenase (DLD) at K320.

High Sugar Induced RCC2 Lactylation Drives Breast Cancer Tumorigenicity Through Upregulating MAD2L1

This study finds that KAT2A mediates the lactylation modification of the non‐histone protein RCC2 at K124, facilitating the recruitment of free SERBP1 and aiding in the formation of the initial SERBP1‐MAD2L1 complex. SERBP1 enhances the stability of MAD2L1 mRNA and promotes cellular proliferation.
