Targeting lysosomal quality control as a therapeutic strategy against aging and diseases

open access: yesMedicinal Research Reviews, Volume 44, Issue 6, Page 2472-2509, November 2024.
Abstract Previously, lysosomes were primarily referred to as the digestive organelles and recycling centers within cells. Recent discoveries have expanded the lysosomal functional scope and revealed their critical roles in nutrient sensing, epigenetic regulation, plasma membrane repair, lipid transport, ion homeostasis, and cellular stress response ...

Tissue invasion and metastasis: molecular, biological and clinical perspectives [PDF]

open access: yes, 2015
Cancer is a key health issue across the world, causing substantial patient morbidity and mortality. Patient prognosis is tightly linked with metastatic dissemination of the disease to distant sites, with metastatic diseases accounting for a vast ...

A Small Molecule, Odanacatib, Inhibits Inflammation and Bone Loss Caused by Endodontic Disease [PDF]

open access: yesInfection and Immunity, 2015
ABSTRACT Periapical disease, an inflammatory disease mainly caused by dental caries, is one of the most prevalent infectious diseases of humans, affecting both children and adults. The infection travels through the root, leading to inflammation, bone destruction, and severe pain for the patient.

Absence of Dap12 and the αvβ3 integrin causes severe osteopetrosis [PDF]

open access: yes, 2014
In vitro, ligand occupancy of αvβ3 integrin induces phosphorylation of Dap12, which is essential for osteoclast function. Like mice deleted of only αvβ3, Dap12(−/−) mice exhibited a slight increase in bone mass, but Dap12(−/−) mice, lacking another ITAM ...

Searching for “Greener” Bioequivalents of CF3 to Lower its Environmental Impact

open access: yesChemistry – A European Journal, Volume 30, Issue 50, September 5, 2024.
Considering concerns on the negative environmental effects of CF3 containing compounds, we searched for “greener” alternatives. We found −I as the best alternative since it is equivalent in all aspects studied, size, electronic and lipophilic, followed by −F, ‐cyclo‐Pr, −2‐oxetyl, −SF5 and −SCH3. We suggest using these functionalities as an alternative

ATHEROSCLEROSIS, VASCULAR CALCIFICATION AND BONE LOSS (OSTEOPOROSIS): COMMON PATHOPHYSIOLOGICAL MRCHANISMS DEVELOPMENT OF THE DESEASES AND RESEARCH NOVEL DRAGS FOR DUAL THERAPIE

open access: yesАтеросклероз, 2016
Atherosclerosis and osteoporosis are public health problems, with several epidemiological links, and they might be related to each other in terms of pathogenesis and therapeutic agents.

Heat induced evaporative antisolvent nanoprecipitation (HIEAN) of itraconazole [PDF]

open access: yes, 2014
YesItraconazole (ITR) is an antifungal drug with a limited bioavailability due to its poor aqueous solubility. In this study, ITR was used to investigate the impact of nanonisation and solid state change on drug’s apparent solubility and dissolution. A

Odanacatib increases mineralized callus during fracture healing in a rabbit ulnar osteotomy model [PDF]

open access: yesJournal of Orthopaedic Research, 2015
ABSTRACTThe effects of the cathepsin K inhibitor odanacatib (ODN) on fracture healing were monitored for ∼6 and 15 weeks post‐fracture in two separate studies using the unilateral transverse mid‐ulnar osteotomy model in skeletally mature female rabbits. Rabbits were pre‐treated for 3–4 weeks with vehicle (Veh), ODN (2 mg/kg, po, daily), or alendronate (

Immune mediated support of metastasis: Implication for bone invasion

open access: yesCancer Communications, Volume 44, Issue 9, Page 967-991, September 2024.
Abstract Bone is a common organ affected by metastasis in various advanced cancers, including lung, breast, prostate, colorectal, and melanoma. Once a patient is diagnosed with bone metastasis, the patient's quality of life and overall survival are significantly reduced owing to a wide range of morbidities and the increasing difficulty of treatment ...

Cyclopropanation of Aryl and Styryl Acetonitriles With Selenium‐Based Dielectrophiles

open access: yesEuropean Journal of Organic Chemistry, Volume 27, Issue 21, June 3, 2024.
Base‐promoted cyclopropanations of aryl or styrylacetonitriles with vinyl selenones are described. The formation of the two new C−C bonds proceeds through a Michael addition followed by a cyclization. Such reactions are usually difficult with pronucleophiles not bearing an additional geminal electron‐withdrawing group.
