Hepatic Lipid Stress Drives Metabolic Dysfunction‐Associated Steatohepatitis Progression through FTO O‐GlcNAcylation‐Induced m6A Demethylation of HRAS

open access: yesAdvanced Science, EarlyView.
Lipid stress drives O‐GlcNAcylation of the m6A demethylase FTO at Ser195, enhancing its stability and demethylase activity. This modification removes m6A from HRAS mRNA, preventing its degradation and activating PI3K/AKT signaling to promote hepatic steatosis and inflammation.

Targeting Macrophage STING O‐GlcNAcylation via Small‐Molecule Allosteric Modulation for Psoriasis Therapy

open access: yesAdvanced Science, EarlyView.
Paeoniflorin allosterically targets the STING cyclic dinucleotide‐binding domain, stabilizes an inactive open conformation, and disrupts STING–OGT interaction to reduce Thr229 O‐GlcNAcylation and downstream signaling. This mechanism restrains myeloid STING activation, alleviates psoriasis‐like inflammation, and is supported by analyses of patient ...

Fc‐Dependent Targeting of the GAS6–MERTK Axis With Monoclonal and Bispecific Antibodies Overcomes Drug Resistance Across EGFR Mutations in Exons 19, 20, and 21

open access: yesAdvanced Science, EarlyView.
EGFR mutations drive a substantial fraction of non‐small cell lung cancers (NSCLC) and are effectively targeted by tyrosine kinase inhibitors (TKIs), including osimertinib. However, resistance inevitably develops through secondary mutations and compensatory receptor activation, including the GAS6 receptor MERTK.

Loss of Intestinal MSTN Promotes GLP‐1 Secretion Through SGLT1 Palmitoylation

open access: yesAdvanced Science, EarlyView.
Intestinal MSTN deficiency enhances glucose‐stimulated GLP‐1 secretion and glucose tolerance in high‐fat diet‐fed mice. Mechanistically, loss of MSTN promotes YY1‐associated ZDHHC20 expression, increasing SGLT1 S‐palmitoylation at Cys301 and its plasma membrane localization in L cells.

NSUN3‐Associated m5C Modification of tRF5‐GlyTCC Suppresses Colorectal Cancer Progression via RPL23‐Linked Nucleolar Stress and p53‐Associated Autophagy and Enhances 5‐FU Sensitivity

open access: yesAdvanced Science, EarlyView.
This study identifies that NSUN3‐mediated m5C modification stabilizes tRF5‐GlyTCC, linking ribosomal homeostasis, nucleolar stress responses, and p53 signaling changes. This regulatory axis suppresses colorectal cancer and enhances chemotherapy response to 5‐FU, positioning tRF5‐GlyTCC as a potential therapeutic target for improving drug sensitivity in
