Novel diazabicycloalkane delta opioid agonists

Bioorganic & Medicinal Chemistry, 2015
Here we report the investigation of diazabicycloalkane cores as potential new scaffolds for the development of novel analogues of the previously reported diazatricyclodecane selective delta (δ) opioid agonists, as conformationally constrained homologues of the reference δ agonist (+)-4-[(αR)-α((2S,5R)-4-allyl-2,5-dimethyl-1-piperazinyl)-3-methoxybenzyl]

Adverse Effects of Opioid Agonists and Agonist-Antagonists in Anaesthesia

Drug Safety, 1998
The traditional view of opioids held that the individual opioid agonists shared the same mechanism of action, differing only in their potency and pharmacokinetic properties. However, recent advances in opioid receptor pharmacology have made this view obsolete.

Antagonistic Modulation Between the Delta Opioid Agonist BW373U86 and the Mu Opioid Agonist Fentanyl in Mice

The Journal of Pharmacology and Experimental Therapeutics, 1997
This study was performed to assess the interactions that occur between delta- and mu opioid receptors by studying effects of the systemically active nonpeptide delta agonist BW373U86 and the mu agonist fentanyl in mice. Concentrations of the compounds were varied, and analgesic responses were determined by 55 degrees C hot-plate assays.

[Opioid Agonist Therapy (OAT) for opioid dependency].

Therapeutische Umschau. Revue therapeutique, 2020
Opioid Agonist Therapy (OAT) for opioid dependency Abstract. The process of evaluating the necessity of an Opioid Agonist Therapy (OAT) is relatively simple. Once the criteria for an opioid dependency according to the ICD-10 (or DSM V) are met, treatment should be offered. Adequate individual dosing of the opioid agonist is crucial to treatment success.

Specific kappa opioid receptor agonists.

Farmaco (Societa chimica italiana : 1989), 2001
The results of studies on the design of a heterocyclic scaffold for the dynorphin A pharmacophore and on structure-affinity relationships in the MPCB/CCB series are described. The representative ligands provide insights to binding modes of benzomorphan derivatives to the kappa opioid receptor.

Recognition of opioid agonist and antagonist in the opioid receptor binding site

Life Sciences, 1982
By treating the rat crude synaptosomal fraction with 5,5'-dithio-bis-(2-nitrobenzoic acid), DTNB, a marked decrease of stereo-specific binding of opioid agonist (dihydromorphine or D-Ala-D-Leu-enkephalin) was observed, but there was no effect in the case of the binding of opioid antagonist (naloxone or diprenorphine).

The Agonist Paradox: Agonists and Antagonists of Acetylcholine Receptors and Opioid Receptors

Chemistry & Biodiversity, 2005
AbstractIn contrast to antagonists, agonists tend to induce considerable conformational changes in their receptors, resulting in opening of ion channels, either directly or via secondary messengers. These conformational transformations require great energy expenses.

Psychotherapeutic Benefits of Opioid Agonist Therapy

Journal of Addictive Diseases, 2008
Opioids have been used for centuries to treat a variety of psychiatric conditions with much success. The so-called "opium cure" lost popularity in the early 1950s with the development of non-addictive tricyclic antidepressants and monoamine oxidase inhibitors. Nonetheless, recent literature supports the potent role of methadone, buprenorphine, tramadol,

Buprenorphine: The Opioid that Cried 'Partial Agonist'

Journal of Opioid Management
Background: Although buprenorphine use has increased dramatically over the past decade, its unique pharmacologic and pharmacokinetic profile often leads to misconceptions about its overall utility and has created a drastic underrepresentation in patients with chronic non-cancer pain.
