Observations on the thermostability of orosomucoid

Summary Orosomucoid is shown to have no special resistance to thermal denaturation, contrary to previous thought. Earlier conclusions probably arose from the great solubility of the molecule, even at elevated temperature. Under otherwise physiological conditions, orosomucoid begins denaturing at ∼46°C with a T m of 62°C.

AbstractStudies of orosomucoid (α1‐acid glycoprotein) in human serum have revealed that orosomucoid is a mixture of molecules with differences in the glycan chains. This microheterogeneity has been studied using crossed affinoimmuno‐electrophoresis with the lectin concanavalin A which binds to biantennary glycans.

Abstract An acidic glycoprotein has been isolated from nephrotic urine by means of DEAE cellulose chromatography. The homogeneity of the glycoprotein was investigated and an antiserum raised to it in rabbits. Periodate oxidation of the glycoprotein was carried out in order to assist in determining the antigenic moiety of the molecule.

Serum orosomucoid concentration in newborn infants

Serum orosomucoid concentration was measured by laser nephelometry in 1970 serum samples collected from 1170 full term and preterm infants. The determinations were carried out in 1 h. Reference values are given: they show that the low levels at birth are influenced by gestational age.

Inhibition of viral hemagglutination by aggregated orosomucoid

Abstract The aggregation of orosomucoid by heating at 80 °, and its resultant capacity to inhibit viral hemagglutination by PR8 influenza virus has been studied. Heating orosomucoid in unbuffered saline resulted in a polydisperse aggregate. Fractionation of such preparations on Sephadex G-200 resulted in a separation into three components.
