Composition Study of Polyphyllin in Paris polyphylla by Ultrasound-Assisted Deep Eutectic Solvent Extraction Combined with UHPLC-MS/MS

open access: yesMolecules
Paris polyphylla (Chonglou), a medicinal herb documented in Shennong’s Classic of Materia Medica and a key component of formulas such as Yunnan Baiyao, is a rare and endangered plant prized for its bioactive steroidal saponins, notably polyphyllin I (PPI)

Analysing the significance of small conformational changes and low occupancy states in serial crystallographic data

open access: yesFEBS Open Bio, EarlyView.
This protocol paper outlines methods to establish the success of a time‐resolved serial crystallographic experiment, by means of statistical analysis of timepoint data in reciprocal space and models in real space. We show how to amplify the signal from excited states to visualise structural changes in successful experiments.

Optimization of High Temperature and Pressurized Steam Modified Wood Fibers for High-Density Polyethylene Matrix Composites Using the Orthogonal Design Method

open access: yesMaterials, 2016
The orthogonal design method was used to determine the optimum conditions for modifying poplar fibers through a high temperature and pressurized steam treatment for the subsequent preparation of wood fiber/high-density polyethylene (HDPE) composites. The

Large‐scale bidirectional arrayed genetic screens identify OXR1 and EMC4 as modifiers of αSynuclein aggregation

open access: yesFEBS Open Bio, EarlyView.
Activation of the mitochondrial protein OXR1 increases pSyn129 αSynuclein aggregation by lowering ATP levels and altering mitochondrial membrane potential, particularly in response to MSA‐derived fibrils. In contrast, ablation of the ER protein EMC4 enhances autophagic flux and lysosomal clearance, broadly reducing α‐synuclein aggregates.

Long‐term hippocampal alterations and cognitive impairment in a murine model of surgical sepsis

open access: yesFEBS Open Bio, EarlyView.
Using a mouse model of surgical sepsis, we tested long‐term memory and analyzed the transcriptome of single cells isolated from the hippocampus. Survivor mice showed worse memory, loss of certain brain cell subpopulations, and abnormal immune cell activity—suggesting that post‐sepsis brain alterations may be linked to cognitive deficits.

Nearly Orthogonal Randomized Designs

open access: yesJournal of the Royal Statistical Society Series B: Statistical Methodology, 1993
SUMMARY A new method of experimental randomization is presented that is suitable under additive general linear models for treatment and unit effects. The method is designed for a standard normal theory analysis of the data. The method is motivated by thinking of the randomization strategy, which may be represented as a distribution on ...
