Outer membrane protein evolution
Current Opinion in Structural Biology, 2021Outer membrane proteins have remarkably homogeneous structure. They are all up down β-barrels. Up down barrels themselves are composed of repeated sets of β-hairpins. The consistency of the usage of the β-hairpin throughout the outer membrane milieu allows for interrogation of the evolution of these repetitive structures.

Biogenesis of mitochondrial outer membrane proteins
Mitochondria are surrounded by two distinct membranes: the outer and the inner membrane. The mitochondrial outer membrane mediates numerous interactions between the mitochondrial metabolic and genetic systems and the rest of the eukaryotic cell. Proteins of this membrane are nuclear-encoded and synthesized as precursor proteins in the cytosol. They are

Insights into outer membrane protein crystallization [PDF]
Outer membrane proteins are structurally distinct from those that reside in the inner membrane and play important roles in bacterial pathogenicity and human metabolism. X-ray crystallography studies on >40 different outer membrane proteins have revealed that the transmembrane portion of these proteins can be constructed from either beta-sheets or less ...

Folding of outer membrane proteins
Archives of Biochemistry and Biophysics, 2013Outer membrane proteins (OMPs) represent a large group of β-barrel proteins found both in the membranes of both bacteria and eukaryotes. Their general ease of expression and refolding and straightforward methods to monitor their degree of folding conspire to make OMPs excellent model systems to investigate how the membrane environment and other ...

Outer Membrane Protein Purification
2003The major outer membrane proteins (OMPs) from Neisseria meningitidis, which are expressed at high levels, are subdivided in five classes based on molecular weight (1,2) (see Table 1). The membrane of Gram-negative bacteria consists of three layers: (i) the cytoplasmic or inner membrane, (ii) the peptidoglycan layer and (iii) the outer membrane (Figure 1). The cytoplasmic membrane consists of a phospholipid bilayer in which proteins are present which can play a role in solute transport, generation of energy and synthesis of cell ...

Biogenesis of Outer Membrane Proteins
2003The cell envelope of Gram-negative bacteria consists of two membranes, the inner membrane, which is a phospholipid bilayer, and the outer membrane, which is an asymmetrical bilayer with phospholipids and lipopolysaccharides (LPS) in the inner and outer monolayers, respectively. The membranes are separated by the peptidoglycan-containing periplasm. Both

Comparison of Chlamydia outer membrane complex to recombinant outer membrane proteins as vaccine
Vaccine, 2020The Chlamydial outer membrane complex (COMC) from the elementary body (EB) is a protein rich insoluble outer membrane shell from which cytosolic proteins have been extracted with detergent. In this study we conducted mass spectrometry experiments to detect proteins in the COMC prepared from C. muridarum EB.

The amino terminal 29 amino acids of the outer mitochondrial membrane protein of yeast, OMM70 (MAS70), consisting of the targeting and membrane anchor domains, has been fused to a reporter protein, dihydrofolate reductase. The hybrid protein, designated pOMD29, was efficiently imported into the outer membrane of rat heart mitochondria by a process ...

Assembly of Bacterial Outer Membrane Proteins
2012Various methods that are routinely used to study the subcellular localization of membrane proteins in wild-type Gram-negative bacteria fall short in genetic studies addressing the biogenesis of outer membrane proteins (OMPs). Here, we describe three biochemical methods that can be used in such studies to evaluate the proper assembly of OMPs into the ...
