PARP Inhibitors plus Anlotinib as Bridging Therapy for Armored CAR‐T Cells in Ovarian Cancer Enhances Infiltration and Antitumor Efficacy

open access: yesAdvanced Science, EarlyView.
This study shows that a PARP inhibitor combined with anlotinib, administered as bridging therapy, preconditions the tumor microenvironment to enhance the infiltration and antitumor activity of subsequently infused CAR‐T cells across preclinical ovarian cancer models, supporting bridging therapy as a promising strategy to address limited CAR‐T cell ...

Ferroptosis Suppression by the M6A Reader IGF2BP1 Underlies Gemcitabine Resistance in Pancreatic Ductal Adenocarcinoma

open access: yesAdvanced Science, EarlyView.
IGF2BP1 promotes gemcitabine resistance in pancreatic cancer by stabilizing m6A‐modified FTH1 transcripts and protecting them in stress granules, thereby suppressing ferroptosis. Pharmacological inhibition of IGF2BP1 disrupts this protective pathway, restores ferroptotic sensitivity, and enhances gemcitabine efficacy in resistant tumors.

VlPAG/DRN Microglia Drive Neuropathic Pain‐Induced Depression via a Defined Neuroimmune Axis

open access: yesAdvanced Science, EarlyView.
Peripheral nerve injury (SNI) induces neuropathic pain, activates midbrain microglia, and triggers an NLRP3–IL‐1β–dependent inflammatory cascade. IL‐1β signals through IL‐1R1 on vlPAG/DRN VGLUT2+ neurons (vlPAG/DRNGlu), inducing hyperexcitability and driving the vlPAG/DRNGlu→BNSTGABA circuit to produce the affective component of neuropathic pain ...

OTUD6A‐Mediated Deubiquitination of PRDX1 Protects Against Oral Ulcer by Restoring Mitochondrial Function

open access: yesAdvanced Science, EarlyView.
In human oral keratinocytes, OTUD6A promoted cell migration by deubiquitinating and stabilizing PRDX1. Conversely, OTUD6A deficiency reduced PRDX1 stability, triggering mitochondrial dysfunction and aggravating OU progression. ABSTRACT Oral ulcers (OU), as the most highly prevalent and recurrent oral mucosal lesion, have an unclear pathogenesis that ...

Livestock Multi‐Omics Integration: A Systematic Framework From Statistical Association to Causal Interpretation

open access: yesAdvanced Science, EarlyView.
A three‐tier livestock multi‐omics framework resolves four typical analytical pitfalls. Moving from statistical association through machine learning preprocessing to triple‐modal causal inference, it converts omics results into genomic selection and gene editing strategies to achieve One Health, underpinned by multi‐omics data, multimodal sequencing ...

EBV‐Driven ODC1 Upregulation Enhances Polyamine Anabolism to Promote Viral Replication and Cisplatin Resistance in Nasopharyngeal Carcinoma

open access: yesAdvanced Science, EarlyView.
EBV‑BZLF1 initiates ODC1‑driven polyamine anabolism that correlates with poor clinical prognosis in nasopharyngeal carcinoma. The ODC1–spermidine axis promotes viral replication, cell proliferation and innate immune evasion. Pharmacological inhibition of ODC1 rescues cisplatin sensitivity, establishing ODC1 as a viable therapeutic target for EBV ...

Toward Predictable Nanomedicine: Current Forecasting Frameworks for Nanoparticle–Biology Interactions

open access: yesAdvanced Intelligent Discovery, EarlyView.
Predictive models successfully screen nanoparticles for toxicity and cellular uptake. Yet, complex biological dynamics and sparse, nonstandardized data limit their accuracy. The field urgently needs integrated artificial intelligence/machine learning, systems biology, and open‐access data protocols to bridge the gap between materials science and safe ...

Cell Segmentation Beyond 2D—A Review of the State‐of‐the‐Art

open access: yesAdvanced Intelligent Discovery, EarlyView.
Cell segmentation underpins many biological image analysis tasks, yet most deep learning methods remain limited to 2D despite the inherently 3D nature of cellular processes. This review surveys segmentation approaches beyond 2D, comparing 2.5D and fully 3D methods, analyzing 31 models and 32 volumetric datasets, and introducing a unified reference ...

Clinically Informed Intelligent Classification of Ovarian Cancer Cells by Label‐Free Holographic Imaging Flow Cytometry

open access: yesAdvanced Intelligent Systems, Volume 7, Issue 3, March 2025.
Quantitative phase maps of single cells recorded in flow cytometry modality feed a hierarchical architecture of machine learning models for the label‐free identification of subtypes of ovarian cancer. The employment of a priori clinical information improves the classification performance, thus emulating the clinical application of liquid biopsy during ...
