Structural insights into an engineered feruloyl esterase with improved MHET degrading properties

open access: yesFEBS Letters, EarlyView.
A feruloyl esterase was engineered to mimic key features of MHETase, enhancing the degradation of PET oligomers. Structural and computational analysis reveal how a point mutation stabilizes the active site and reshapes the binding cleft, expading substrate scope.

Oscillation for advanced differential equations with oscillating coefficients

open access: yesInternational Journal of Mathematics and Mathematical Sciences, 2003
Some sufficient conditions are established for the oscillation of all solutions of the advanced differential equation x′(t)−p(t)x(t+τ)=0, t≥t0, where the coefficient p(t)∈C([t0,∞),R) is oscillatory, and τ is a positive constant.

Valosin‐containing protein counteracts ATP‐driven dissolution of FUS condensates through its ATPase activity in vitro

open access: yesFEBS Letters, EarlyView.
Biomolecular condensates formed by fused in sarcoma (FUS) are dissolved by high ATP concentrations yet persist in cells. Using a reconstituted system, we demonstrate that valosin‐containing protein (VCP), an AAA+ ATPase, counteracts ATP‐driven dissolution of FUS condensates through its D2 ATPase activity.

Hyperosmotic stress induces PARP1‐mediated HPF1‐dependent mono(ADP‐ribosyl)ation

open access: yesFEBS Letters, EarlyView.
Sorbitol‐induced hyperosmotic stress rapidly induces reversible mono(ADP‐ribosyl)ation (MARylation) on PARP1 without the signs of genotoxic signaling. We show that PARP1 autoMARylation is HPF1 dependent and forms hydroxylamine‐resistant O‐glycosidic linkages.

An isoform of 14‐3‐3 protein regulates transbilayer lipid movement at the plasma membrane

open access: yesFEBS Letters, EarlyView.
Loss of 14‐3‐3ζ in CHO cells confers resistance to exogenous phosphatidylserine (PS) and impairs endocytosis‐independent inward flip‐flop of fluorescent PS at the plasma membrane. RNAi‐mediated knockdown reproduces this defect, while no additive effect is seen in ATP11C‐deficient cells.

The ubiquitin ligase RNF115 is required for the clearance of damaged lysosomes

open access: yesFEBS Letters, EarlyView.
Upon lysosomal rupture, an E3 ubiquitin ligase RNF115 translocates from the cytosol to the damaged lysosomal membrane. Moreover, RNF115 depletion impairs the clearance of damaged lysosomes, identifying it as a key regulator of lysosomal quality control.

Positive solutions for a second-order \Phi-Laplacian equations with limiting nonlocal boundary conditions

open access: yesElectronic Journal of Differential Equations, 2016
Motivated, mainly, by the works of Fewster-Young and Tisdell [9,10] and Orpel [30], as well as the papers by Karakostas [21,22,23], we give sufficient conditions to guarantee the existence of (nontrivial) solutions of the second-order Phi-Laplacian

pH‐mediated activation of the lysosomal arginine sensor SLC38A9

open access: yesFEBS Letters, EarlyView.
Cells monitor nutrient levels via the lysosomal transporter SLC38A9 to activate the mechanistic target of rapamycin complex 1 (mTORC1). This study reveals that SLC38A9 function is regulated by pH. We identified histidine 544 as a critical pH sensor that undergoes conformational changes to control amino acid efflux from lysosomes; therefore, it ...

Dissolution of Diamond in Water–Chloride Fluids at Mantle P-T Conditions

open access: yesMinerals
Syngenetic fluid inclusions in natural diamonds are indicators of the composition of fluids responsible for growth and crystallization conditions. The chloride concentration in saline fluid inclusions of natural diamonds reaches 50 wt%. We study the dissolution of diamonds in the H2O-KCl-NaCl system at temperatures of 1200 °C and 1400 °C and a pressure

Quantifying the P–T conditions of silica enrichment in Archaean cratonic spinel-facies peridotites

open access: yesMineralogical Magazine
Abstract Anomalously high amounts of orthopyroxene are an unexpected feature of some depleted peridotite xenoliths from the cratonic lithosphere. This investigation presents new petrographic, geochemical and thermodynamic modelling data for silica-rich spinel harzburgites and one garnet–spinel harzburgite from the Kaapvaal Craton ...
