Appearance and development of P700 oxidation and photosystem I activity in etio-chloroplasts prepared from greening barley leaves

The development of photosystem I activity of plastids isolated from greening barley (Hordeum distichum, L.) leaves was studied. The electron transport activity in photosystem I was measured as anthraquinone-mediated oxygen uptake and as light induced absorbance changes of the reaction centre molecule P700.

[Kinetics of photoinduced oxidative-reductive transformation of the reaction center P700 in leaves of C3- and C4-plants].

The light-induced changes in the kinetics of the oxidation of photosystem I reaction center P700 in leaves of C3 and C4 plants, depending on the time of leaf adaptation to dark before the illumination were studied. The EPR 1 signal from oxidized centers P+700 was used to monitor the kinetics of P700 redox transients.

Diverse strategies in P700 oxidation system for suppressing ROS production were acquired during the evolution of photosynthetic organisms

We developed the method to produce ROS intrinsically in photosynthetic organisms: repetitive short-pulse (rSP) illumination. rSP illumination fulfills electrons in photosynthetic electron transport (PET) system, where O2 is reduced to superoxide radical and O2 is also excited to singlet O2 at photosystem I (PSI) of PET system.

A sharp fall of ESR signal I is observed at temperature decrease while switching on light of 600 nm on the background of light of 707 nm; kinetics of the initiation of the signal under the illumination of light of 707 nm gets more complicated; the induction phase induced by previous illumination with the light of 600 nm increases.
