Wang et al. uncover metabolic regulation of antiviral immunity through CMPK2 palmitoylation, which regulates CMPK2 mitochondrial localization and is promoted by ZDHHC20 but reversed by PPT1, inhibition of which antagonizes viral infection in mice.
