Tumor‐Derived LAMB3 Drives Immunosuppressive LRRC15+ Fibroblast Formation During Pancreatic Ductal Adenocarcinoma Development

A single‐cell atlas of pancreatic ductal adenocarcinoma development reveals progressive ductal‐fibroblast‐immune crosstalk. Tumor‐derived LAMB3 drives the formation of immunosuppressive LRRC15+ fibroblasts through the ITGB1/FAK/MAPK/FOSL2 signaling. Tumor‐Intrinsic ARHGEF3 Enhances Antitumor Immunity by Promoting T‐Cell Infiltration and Limiting Myeloid Cell‐Mediated Immunosuppression

ARHGEF3 is broadly downregulated across human cancers and correlates with patient prognosis. Tumor‐intrinsic ARHGEF3 activates the RHOA–ROCK–PTEN cascade to inhibit AKT signaling, thereby promoting chemokine‐driven T‐cell infiltration and relieving lipid‐mediated myeloid immunosuppression.

MiR‐940 Suppresses Ferroptosis by Controlling Expression of Key Regulatory Genes

A CRISPR‐based screening identified miR‐940 as a critical suppressor of ferroptosis in cancer. By coordinating the downregulation of pro‐ferroptotic genes with the upregulation of GPX4, miR‐940 establishes a regulatory network that protects against ferroptosis and correlates with poor clinical outcomes in distinct cancer entities.
