Here, hydrogen‐deuterium exchange mass spectrometry can show and reveal gluing mechanisms that vary between protein binding partners. A supramolecular fluorescent probe, TPE‐4B/4Q[7], was designed to target lysosomes. The restriction of intramolecular rotation effect mediated by Q[7] enhances the fluorescence of TPE‐4B/4Q[7], enabling selective detection of Fe3+ with a detection limit of 1.23 × 10⁻⁶ M.
