Macrophage‐derived MLKL in alcohol‐associated liver disease: Regulation of phagocytosis
Hepatology, EarlyView., 2022 EtOH causes leaky gut allowing bacteria and PAMPs into the liver, resulting in hepatic inflammation and injury. We demonstrate that LPS induces STAT1‐mediated expression and phosphorylation of MLKL in macrophages and identify a novel function that myeloid MLKL translocates to phagosomes and lysosomes and regulates phagocytosis, which contributes to the Xiaoqin Wu, Xiude Fan, Megan R. Advanced Science, EarlyView.ABSTRACT Background
Therapeutic resistance limits durable survival in advanced/metastatic renal cell carcinoma (RCC) treated with first‐line tyrosine kinase inhibitor (TKI) plus immune checkpoint inhibitor (ICI). We sought to define key resistance drivers and actionable targets.Jinchen Luo, Hansen Lin, Haoqian Feng, Lei Tan, Xi Liu, Yong Huang, Junjie Cen, Jiajie Chen, Xinwei Zhou, Mingjie Lin, Wuyuan Liao, Zheyu Ai, Minyu Chen, Yinghan Wang, Wei Chen, Junhang Luo, Yanping Liang +16 morewiley +1 more sourceGene Correction Enhances Dopaminergic Cell Therapy in a Nonhuman Primate Model of Parkinson's Disease
Advanced Science, EarlyView.LRRK2‐mutant induced pluripotent stem cells (iPSCs) were derived from a patient with Parkinson's disease (PD). Using CRISPR/Cas9–mediated gene editing, the pathogenic LRRK2 mutations were precisely corrected, and isogenic dopaminergic neural progenitor cells (DA‐NPCs) were subsequently generated.Qing Yan, Chongchong Xu, Jiangmei Gao, Pu Wang, Qingling Wu, Ying Jin, Binyan Lu, Mu Li, Mingting Shao, Bihai Li, Zhenghui Su, Yijng Zhang, Jianhuan Chen, Haojie An, Mengyao Huang, Xiaoji Zhuang, Yuhui Shen, Fenglin Wang, Nana Xu, Yiyan Liu, Lei Tang, Xiuling Zhong, Minyan Zhong, Jiecong Chen, Zhenhuang Wang, Xingrong Luo, Sheng Liu, RuiFeng Liu, Ling Zhang, Junhua Rao +29 morewiley +1 more sourceA Spatially Directed Microneedle Patch Enables Intratumoral Co‐Delivery of FOLFIRINOX, Surufatinib, and Anti‐PD‐1 for Chemo‐Immunotherapy of Pancreatic Ductal Adenocarcinoma
Advanced Science, EarlyView.A double‐layered shell‐core microneedle patch is developed to co‐deliver FOLFIRINOX, surufatinib, and anti‐PD‐1 for localized chemo‐immunotherapy of PDAC. This strategy achieves sustained tumor suppression, reduces metastasis, and reprograms the TME by enhancing CD8+ T‐cell infiltration and inhibiting Tregs and M2 macrophages infiltration, while ...Tingting Kong, Ximo Xu, Xiao Zhang, Chuntao Wu, Zhengjun Qiu, Beiyuan Hu, Zihao Qi, Qiang Tian, Yuqin Yang, Hanguang Dong, Fei Wu, Tuo Jin, Yan Zheng, Jiang Long +13 morewiley +1 more sourceDNA‐mimic for Specific Surface Functionalization of Zr‐MOFs for Bacterial Targeting

American Journal of Medical Genetics Part A, EarlyView.ABSTRACT
TECPR2 is a key regulator of autophagy, encoded by the TECPR2 gene. Pathogenic variants in this gene have been linked to a rare hereditary sensory and autonomic neuropathy with intellectual disability (HSAN9). We report a teenage female with a syndromic intellectual disability disorder associated with neuromuscular abnormalities.Teresa Zhao, Andrew P. Fennell, Tanavi Sharma, Katrina M.
