Adaptaquin is selectively toxic to glioma stem cells through disruption of iron and cholesterol metabolism

open access: yesMolecular Oncology, EarlyView.
Adaptaquin selectively kills glioma stem cells while sparing differentiated brain cells. Transcriptomic and proteomic analyses show Adaptaquin disrupts iron and cholesterol homeostasis, with iron chelation amplifying cytotoxicity via cholesterol depletion, mitochondrial dysfunction, and elevated reactive oxygen species.

Patient‐specific pharmacogenomics demonstrates xCT as predictive therapeutic target in colon cancer with possible implications in tumor connectivity

open access: yesMolecular Oncology, EarlyView.
This study integrates transcriptomic profiling of matched tumor and healthy tissues from 32 colorectal cancer patients with functional validation in patient‐derived organoids, revealing dysregulated metabolic programs driven by overexpressed xCT (SLC7A11) and SLC3A2, identifying an oncogenic cystine/glutamate transporter signature linked to ...

Locked nucleic acid inhibits amplification of contaminating DNA in real-time PCR

open access: yesBioTechniques, 2005
Locked nucleic acid (LNA®) is a modified DNA with increased binding affinity for complementary DNA sequences. Our strategy was to use this property of LNA to inhibit undesired PCR amplification (e.g., from contaminating genomic DNA) in a cDNA-based assay.

Predictors of response and rational combinations for the novel MCL‐1 inhibitor MIK665 in acute myeloid leukemia

open access: yesMolecular Oncology, EarlyView.
This study characterizes the responses of primary acute myeloid leukemia (AML) patient samples to the MCL‐1 inhibitor MIK665. The results revealed that monocytic differentiation is associated with MIK665 sensitivity. Conversely, elevated ABCB1 expression is a potential biomarker of resistance to the treatment, which can be overcome by the combination ...

Molecular diagnosis by PCR-DHPLC technique of wood-decay fungi in historical buildings in Italy [PDF]

open access: yes, 2012
Wood inhabiting fungi cause real problems in the preservation of wooden surfaces and are responsible for the deterioration of cultural heritage. The identification of fungi based on morphological characteristics are still a topical issue.

Species-specific Real Time-PCR primers/probe systems to identify fish parasites of the genera Anisakis, Pseudoterranova and Hysterothylacium (Nematoda: Ascaridoidea) [PDF]

open access: yes, 2018
Ascaridoid nematodes belonging to the genera Anisakis and Pseudoterranova are heteroxenous parasites, involving marine mammals as definitive hosts in their life-cycles, whereas crustaceans (krill), fish and squids acting as intermediate/paratenic hosts.

Feasibility of a ctDNA multigenic panel for non‐small‐cell lung cancer early detection and disease surveillance

open access: yesMolecular Oncology, EarlyView.
Plasma‐based detection of actionable mutations is a promising approach in lung cancer management. Analysis of ctDNA with a multigene NGS panel identified TP53, KRAS, and EGFR as the most frequently altered, with TP53 and KRAS in treatment‐naïve patients and TP53 and EGFR in previously treated patients.

5′-Degenerate 3′-Dideoxy-Terminated Competitors of PCR Primers Increase Specificity of Amplification

open access: yesBioTechniques, 1998
Amplification of a product in PCR with specific primers may be viewed as an artificial Darwinian-type “selection of the fittest”. In other selective systems, such as general evolution, immune system and probably brain cortex, the stringency of selection ...

Capturing the ‘ome’ : the expanding molecular toolbox for RNA and DNA library construction [PDF]

open access: yes, 2018
All sequencing experiments and most functional genomics screens rely on the generation of libraries to comprehensively capture pools of targeted sequences.
