A synthetic peptide substrate for initiation factor-2 kinases

A peptide P(45-56) corresponding to residues 45-56 (sequence: ILLSELSRRRIR) of eIF-2 alpha was synthesised. It was phosphorylated by both of the well characterised eIF-2 alpha kinases viz.; the heme-controlled repressor (HCR) and the double stranded RNA-dependent inhibitor (dsI).

Antimicrobial Peptide Buforin I Inhibits Tissue Factor-Initiated Coagulation

The enhanced extrinsic blood coagulation following septic shock often manifests cardiovascular complications. The upregulated monocytic tissue factor (mTF) was shown to be a primary contributor to the extrinsic hypercoagulation following acute bacterial endotoxin (LPS) infection. A single-stage clotting assay monitors TF-initiated coagulation.

Summary In a cell-free amino acid incorporating system using preincubated reticulocyte ribosomes, the transfer of methionine from [35S] Met-tRNAfMet in response to poly r((U-G) messenger (in the presence of unlabeled valine, cysteine and crude tRNA) was dependent on addition of the 0.5M KCl wash of crude reticulocyte ribosomes.

Guanosine triphosphate - dependent autocatalytic phosphorylation of peptide initiation factor 2 from rat liver

If incubated with [γ-32P]guanosine triphosphate, homogenous peptide initiation factor 2 from rat liver liberates the terminal phosphate from GTP and utilizes 25–40% of it for its own phosphorylation. This phosphorylation is apparently autocatalytic since it does not require any additional proteins.

Abstract A protein factor, obtained from 0.5 M KCl extracts of rabbit reticulocyte ribosomes, forms a GTP-dependent complex with the initiator tRNA from rabbit liver, Met-tRNAMetf. This activity has been purified approx. 125-fold by ion-exchange chromatography upon DEAE-cellulose and hydroxylapatite.
