Peroxynitrite: A Putative Cytotoxin

Pharmacology & Toxicology, 1998
Abstract:In recent years it has become apparent that peroxynitrite. which is one of the toxic metabolites originating from the reaction of nitric oxide and superoxide presents a number of pathologic states in which free radicals are thought to be involved.

Peroxynitrite Scavenging by Flavonoids

Biochemical and Biophysical Research Communications, 1997
The peroxynitrite scavenging activity of a series of structurally related flavonoids was tested. It was found that flavonoids are excellent scavengers of peroxynitrite. Compared to the known peroxynitrite scavenger ebselen, the most active flavonoids proved to be 10 times more effective.

Peroxynitrite—An ugly biofactor?

BioFactors, 2010
AbstractCellular damage occurring under oxidative conditions has been ascribed mainly to the formation of peroxynitrite (ONOOH/ONOO−) that originates from the reaction of NO•with O2•−. The detrimental effects of peroxynitrite are exacerbated by the reaction with CO2that leads to ONOOC(O)O−, which further decays to the strong oxidant radicals NO2•and ...

Bactericidal activity of peroxynitrite

Archives of Biochemistry and Biophysics, 1992
Peroxynitrite is a strong oxidant formed by macrophages and potentially by other cells that produce nitric oxide and superoxide. Peroxynitrite was highly bactericidal, killing Escherichia coli in direct proportion to its concentration with an LD50 of 250 microM at 37 degrees C in potassium phosphate, pH 7.4.

Peroxynitrite-Dependent Tryptophan Nitration

Chemical Research in Toxicology, 1996
Peroxynitrite (ONOO-), the reaction product of superoxide (O2.-) and nitric oxide (.NO), nitrates tyrosine and other phenolics. We report herein that tryptophan is also nitrated by peroxynitrite in the absence of transition metals to one predominant isomer of nitrotryptophan, as determined from spectral characteristics and liquid chromatography-mass ...

Provoking Neuroprotection by Peroxynitrite

Current Pharmaceutical Design, 2004
Peroxynitrite, the product of the reaction between nitric oxide and superoxide, is spontaneouly formed within most mammalian cells under physiological conditions. Initial work addressing the pathophysiology of peroxynitrite afforded the generally accepted notion that this compound would be the long-term neurotoxic nitric oxide-derivative. However, over

Propofol Is a Peroxynitrite Scavenger

Anesthesia & Analgesia, 1997
P eroxynitrite is a potent oxidant formed by the rapid reaction between nitric oxide and superoxide radical (1). The peroxynitrite anion is relatively stable, but it can rapidly protonate to peroxynitrous acid, an unstable species, which decomposes with a half-life of about one second at pH 7.4 (1,2).

Peroxynitrite and fibrinolytic system: The effect of peroxynitrite on plasmin activity

Molecular and Cellular Biochemistry, 2004
We have shown that peroxynitrite (ONOO-) inhibits streptokinase-induced conversion of plasminogen to plasmin in a concentration-dependent manner and reduces both amidolytic (IC5o approximately 280 microM at 10 microM concentration of enzyme) and proteolytic activity of plasmin.

Peroxynitrite Scavenging Activity of Indole Derivatives: Interaction of Indoles with Peroxynitrite

Journal of Medicinal Food, 2004
One of the products of nitrogen-derived free radicals, peroxynitrite (ONOO(-)), is formed by the reaction of superoxide anion (O(2)(*-)) with nitric oxide (NO). ONOO(-) can cause damage to proteins and DNA through nitration. In particular, proteins and their constituent amino acids have been proven to be extremely sensitive to ONOO(-).
