Phenoxazinone synthase: what's in a name?
Trends in Biotechnology, 2009The name phenoxazinone synthase (PHS, 2-aminophenol:oxygen oxidoreductase, EC 1.10.3.4) is used for the enzyme catalysing the oxidative coupling of substituted o-aminophenols to produce phenoxazinones. This review reveals that the traditional classification of PHS conflicts with recent sequence-based information that shows its relationship with two ...

The copper(II) complexes [Cu(L)NO3] (1-9) of newer N3O ligands (L1-L9) have been synthesized and characterized. The molecular structure of 1, 4, and 7 exhibited nearly a perfect square pyramidal geometry (τ, 0.04-0.11). The Cu-OPhenolate bonds (~ 1.91 Å) are shorter than the Cu-N bonds (~ 2.06 Å) due to the stronger coordination of anionic phenolate ...

AbstractTo study the mechanism of the well known enzymatic oxidation of o‐aminophenols to give phenoxazines (actinomycin, e.g.), substituted o‐aminophenols (I), (III), and (V) which block the reaction at different intermediate stages are examined (see scheme).

The coordination chemistry of the tridentate ligand N-(2-hydroxy-3,5-di-tert-butylphenyl)-2-aminobenzylalcohol H3L has been studied with the copper(II) ion. The ligand is noninnocent in the sense that it is readily oxidized in the presence of air to its o-iminobenzosemiquinonato [L*]2- radical form.

[68] Phenoxazinone synthase (Streptomyces antibioticus)—Syntheses of cinnabarinic acid and actinocin
1970Publisher Summary This chapter describes the assay, purification, and properties of phenoxazinone synthase. Phenoxazinone synthase is possibly involved in the synthesis of actinocin (2-amino-4,6-dimethyl-3-phenoxazinone-1,9-dicarboxylic acid), the chromophore of actinomycin.

Crystallization and initial X-ray analysis of phenoxazinone synthase fromStreptomyces antibioticus
Acta Crystallographica Section D Biological Crystallography, 2004Phenoxazinone synthase, an oligomeric multicopper oxidase produced by Streptomyces antibioticus, is responsible for the six-electron oxidative coupling of two molecules of 4-methyl 3-hydroxyanthraniloyl pentapeptide to form the phenoxazinone chromophore of the antineoplastic agent actinomycin D. Spectroscopic studies have shown that the enzyme contains

[69] Phenoxazinone synthase (Tecoma stans and rat liver)—Synthesis of cinnabarinic acid
1970Publisher Summary This chapter discusses the assay, purification, and properties of phenoxazinone synthase. This convenient and sensitive assay is based on the spectrophotometric estimation of the amount of cinnabarinic acid formed in the reaction mixtures. 3-Hydroxyanthranilic acid is the only known substrate for the Tecoma enzyme.

Particulate fractions from the heads of Drosophila melanogaster catalyze the conversion of o-aminophenols to phenoxazinones. This particulate enzyme is stimulated by Mn2+. It has a number of features which distinguish it clearly from the Mn2+-dependent activity found in the soluble fraction.
