2‐Oxoglutarate Analog‐Based Biomolecular Tools for Exploring Structure–Activity Relationships in Nonheme Iron Enzymes

2‐oxoglutarate (2OG) analogs are rationally designed to elucidate structure–activity relationships (SARs) in 2OG‐dependent nonheme iron (NHFe) enzymes. These analogs are tested against prolyl hydroxylase domain 2 (PHD2). Activity assays reveal that certain analogs compete with 2OG for the PHD2 active site.

Selective Lysine Ubiquitination Using Activated Phenol Esters

Ubiquitination of both peptides and proteins using a novel water‐soluble acylation reagent, based on a 2,4‐dichloro‐6‐sulfonic acid phenol ester, is presented. At alkaline pH, fast lysine ubiquitination is observed on model peptide FLYRANK, and its regioselectivity is confirmed using controls.

Autonomous cells are engineered biological systems capable of biosynthesising and directly incorporating non‐canonical amino acids (ncAAs) into proteins. These systems have the potential to extend the applicability of the genetic code to enable large‐scale fermentative production of proteins carrying ncAAs.
