Extended‐Release Oral Milrinone for the Treatment of Heart Failure With Preserved Ejection Fraction
Background Heart failure with preserved ejection fraction (HFpEF) is an increasingly prevalent form of heart failure, representing half of the total burden of heart failure. We hypothesised that modulation of the phosphodiesterase type 3/cyclic AMP using

3′:5′‐Cyclic Nucleotide Phosphodiesterases in Rat Tissues [PDF]
The rates of enzymatic hydrolysis of cyclic AMP, cyclic GMP, cyclic IMP, cyclic UMP, cyclic CMP and cyclic dTMP (all 3′:5′‐phosphates) have been assayed in several rat tissues. Subcellular distribution studies indicated that, in all tissues except brain and heart, most of the enzymatic activity is confined to the soluble cell fraction. The purine cyclic

A (2R,3R)‐butanediol dehydrogenase from Bacillus subtilis (BsBDH) is engineered for the enantioselective synthesis of 2‐hydroxycyclohexanone. A PASS computational design strategy is proposed to enhance the thermostability of BsBDH. Moreover, ESM‐1v combined with ISM is utilized for enhancing and inverting its stereoselectivity.

Comparison of [11C]TZ1964B and [18F]MNI659 for PET imaging brain PDE10A in nonhuman primates [PDF]
Phosphodiesterase 10A (PDE10A) inhibitors show therapeutic effects for diseases with striatal pathology. PET radiotracers have been developed to quantify in vivo PDE10A levels and target engagement for therapeutic interventions. The aim of this study was

Decoding Human Placental Cellular and Molecular Responses to Obesity and Fetal Growth
Women with obesity often deliver large‐for‐gestational‐age (LGA) infants. Single‐nucleus RNA sequencing of term placenta reveals that hypoxia and TNF‐α signaling in syncytiotrophoblasts are featured in maternal obesity, but inflammatory signatures in Hofbauer cells and response to lipid or carbohydrate metabolism in fibroblasts are specific to LGA.

NAD⁺ homeostasis maintains neuronal integrity through opposing actions of NMNAT2 and SARM1. Loss of NMNAT2 in glutamatergic neurons reprograms cortical metabolism from glucose to lipid catabolism, depletes lipid stores, and triggers inflammation and neurodegeneration.
