Regulation of the levels of phosphoenolpyruvate carboxykinase (GTP) mRNA in rat liver by insulin and glucagon

As can be seen from the data presented in this chapter, glucagon, epinephrine and Bt2cAMP can act directly on isolated rat hepatocytes to markedly increase the concentration of P-enolpyruvate carboxykinase mRNA. The effects of these hormones on enzyme mRNa are rapid, causing a 2–3-fold increase in 90 min of incubation.

Phosphoenolpyruvate carboxykinase (PEPCK) is well known as a key enzyme involved in the metabolic pathway of gluconeogenesis in organisms, but the information about its involvement in immune response is still very limited. The first non-substrate like inhibitors of human cytosolic phosphoenolpyruvate carboxykinase (PEPCK) competitive with GTP are reported. An effort to discover orally active compounds that improve glucose homeostasis in Type 2 diabetics by reversibly inhibiting PEPCK led to the discovery of 1-allyl-3-butyl-8-methylxanthine (5).

The effect of 3-mercaptopicolinic acid on phosphoenolpyruvate carboxykinase (GTP) in the rat and guinea pig

Abstract In vitro , 3-mercaptopicolinic acid inhibited phosphoenolpyruvate carboxykinase activity in supernatant fractions of liver, kidney cortex, and adipose tissue obtained from fasted rats. 3-Mercaptopicolinic acid also inhibited enzymatic activity in the mitochondrial and supernatant fractions of liver obtained from fasted guinea pigs.

Thyroid hormone regulates transcription of the gene for cytosolic phosphoenolpyruvate carboxykinase (GTP) in rat liver

Using an in vitro assay with isolated rat nuclei, we have determined that thyroid hormone causes a 4-6-fold increase in the synthesis of mRNA coding for phosphoenolpyruvate carboxykinase. Proportional changes were seen in the steady-state cytosolic mRNA levels for phosphoenolpyruvate carboxykinase.

Crystal structure of human cytosolic phosphoenolpyruvate carboxykinase reveals a new GTP-binding site

We report crystal structures of the human enzyme phosphoenolpyruvate carboxykinase (PEPCK) with and without bound substrates. These structures are the first to be determined for a GTP-dependent PEPCK, and provide the first view of a novel GTP-binding site unique to the GTP-dependent PEPCK family.
