New techniques for membrane protein crystallization tested on photosystem II core complex of Pisum sativum

Photosynthesis Research, 2007
The crystallization of a given protein is a hard task being even more complicated when the protein shows a hydrophobic behavior. In the case of photosynthetic proteins, the difficulty of the experiments increased due to the high light sensitivity.

Vectorial Charge Transfer Reactions in the Protein-Pigment Complex of Photosystem II

2017
The pigment-protein complex of photosystem II localized in the thylakoid membranes of higher plants, algae, and cyanobacteria is the main source of oxygen on Earth. The light-induced functioning of photosystem II is directly linked to electron and proton transfer across the membrane, which results in the formation of transmembrane electric potential ...

A study of Photosystem II fluorescence emission in terms of the antenna chlorophyll-protein complexes

Biochimica et Biophysica Acta (BBA) - Bioenergetics, 1993
Abstract Fluorescence emission spectra for the seven chlorophyll-protein complexes comprising the antenna system of Photosystem II (PS II) have been measured. All four outer antenna complexes (LHC II, CP24, CP26, CP29) have relatively greater emission near 648 nm and 680 nm with respect to the inner antenna complexes (CP43, CP47, D1/D2/cyt b-559 ...

Protein film voltammetry and co-factor electron transfer dynamics in spinach photosystem II core complex

Photosynthesis Research, 2013
Direct protein film voltammetry (PFV) was used to investigate the redox properties of the photosystem II (PSII) core complex from spinach. The complex was isolated using an improved protocol not used previously for PFV. The PSII core complex had high oxygen-evolving capacity and was incorporated into thin lipid and polyion films.

The effect of norflurazon on protein composition and chlorophyll organization in pigment–protein complex of Photosystem II

Photosynthesis Research, 2005
The pyridazinone-type herbicide norflurazon SAN 9789 inhibiting the biosynthesis of long-chain carotenoids results in significant decrease in PS II core complexes and content of light-harvesting complex (LHC) polypeptides in the 29.5-21 kDa region. The Chl a forms at 668, 676, and 690 nm that belong to LHC and antenna part of PS I disappear completely ...

Isolation of Photosystem II-Enriched Membranes and the Oxygen-Evolving Complex Subunit Proteins From Higher Plants

2004
We describe methods to isolate highly active oxygen-evolving photosystem II (PSII) membranes and core complexes from higher plants, and to purify subunits of the oxygen-evolving complex (OEC). The membrane samples used as the material for various in vitro studies of PSII are prepared by solubilizing thylakoid membranes with the nonionic detergent ...
