Abstract 4-(5',6'-Dimethoxybenzothiazolyl)phthalhydrazide (DBPH) was synthesized as a new, highly sensitive chemiluminescent reagent. The reagent consists of phthalhydrazide and benzothiazole moieties and generates strong chemiluminescence by reaction with hydrogen peroxide in the presence of potassium hexacyanoferrate(III) or peroxidase.

Mechanism for the chemiluminescent photooxidation of phthalhydrazides

The rate constants were determined for the oxidation of phthalhydrazides sensitized by methylene blue, and a mechanism was proposed. The key role of the Superoxide ion in the generation of chemiluminescence was confirmed. The reaction of the methylene blue radical by oxygen was shown to be the major channel for the generation of the superoxide ion.

Theoretical calculation pKa values of phthalhydrazide derivatives in its aqueous solutions

The pKa values of some phthalhydrazide derivatives were calculated using ab initio and two density functional methods to obtain information about their reaction conditions. The results showed that pKa1 was influenced by structural modification as it varied between 0.47 and 6.99, and that pKa2 was relatively less variable from 16.66 to 21.53.
