Abstract Mitochondria from mung bean hypocotyls and potato tubers have been separated from contaminating organelles and membrane fragments on discontinuous sucrose gradients. After removal from the centrifuge tubes only intact mitochondria survive dilution to 0.3 M sucrose.

Protoplasts: Preparation from Higher Plants

Protoplasts can be readily prepared from higher-plant tissue with a concentrated Myrothecium cellulase preparation. An ionic osmoticum produced a higher yield and more viable protoplasts than did the nonionic mannitol osmoticum. Addition of polyuronidases or of ethylenediaminetetra-acetate did not increase the yield.
