Isolation of a Methylocystis strain containing a novel pmoA-like gene [PDF]

open access: hybridFEMS Microbiology Ecology, 2002
Abstract A type II methanotrophic bacterium (Methylocystis strain SC2) was isolated from a polluted aquifer and identified based on morphology and on 16S rRNA gene phylogeny. Primers targeting the particulate methane monooxygenase subunit A gene (pmoA) were used to obtain a PCR product from DNA extract of strain SC2.

Detection of methanotrophs with highly divergent pmoA genes from Arctic soils [PDF]

open access: bronzeFEMS Microbiology Letters, 2002
Tundra soil samples from the Canadian Arctic community, Kuujjuaq, were analyzed for the presence of the soluble (sMMO) and particulate (pMMO) methane monooxygenase genes. Total genomic DNA extracted from these soils was used as template for PCR using sMMO- and pMMO-specific primers, mmoX1-mmoX2 and A189-A682, respectively.

Quantitative Detection of Methanotrophs in Soil by Novel pmoA -Targeted Real-Time PCR Assays [PDF]

open access: greenApplied and Environmental Microbiology, 2003
ABSTRACT Methane oxidation in soils is mostly accomplished by methanotrophic bacteria. Little is known about the abundance of methanotrophs in soils, since quantification by cultivation and microscopic techniques is cumbersome. Comparison of 16S ribosomal DNA and pmoA (α subunit of the particulate methane ...

PMOA-TTS: Introducing the PubMed Open Access Textual Times Series Corpus [PDF]

open access: green
Understanding temporal dynamics in clinical narratives is essential for modeling patient trajectories, yet large-scale temporally annotated resources remain limited. We present PMOA-TTS, the first openly available dataset of 124,699 PubMed Open Access (PMOA) case reports, each converted into structured (event, time) timelines via a scalable LLM-based ...

pmoA Primers for Detection of Anaerobic Methanotrophs [PDF]

open access: greenApplied and Environmental Microbiology, 2011
ABSTRACT Published pmoA primers do not match the pmoA sequence of “ Candidatus Methylomirabilis oxyfera,” a bacterium that performs nitrite-dependent anaerobic methane oxidation. Therefore, new pmoA primers for the detection of “

pMOA 189F-682R v1 [PDF]

open access: gold, 2018
A PCR assay targeting the alpha subunit of the particulate methane monooxygenase gene (pmoA). This is a general assay for methane oxidising bacteria using primers 189F–682R. Note: these primers might occasionally also amplify the amoA gene of ammonia-oxidising bacteria. 189F GGN GAC TGG GAC TTC TGG (target seq. 189 - 207) Holmes et al.

Potential of pmoA Amplicon Pyrosequencing for Methanotroph Diversity Studies [PDF]

open access: greenApplied and Environmental Microbiology, 2011
ABSTRACT We analyzed the potential of pmoA amplicon pyrosequencing compared to that of Sanger sequencing with paddy soils as a model environment. We defined operational taxonomic unit (OTU) cutoff values of 7% and 18%, reflecting methanotrophic species and major phylogenetic pmoA ...

The particulate methane monooxygenase gene pmoA and its use as a functional gene probe for methanotrophs [PDF]

open access: bronzeFEMS Microbiology Letters, 1997
The particulate methane monooxygenase gene pmoA, encoding the 27 kDa polypeptide of the membrane-bound particulate methane monooxygenase, was amplified by PCR from DNA isolated from a blanket peat bog and from enrichment cultures established, from the same environment, using methane as sole carbon and energy source.

pmoA gene reference database (fasta-formatted sequences and taxonomy) [PDF]

open access: greenArchives of Microbiology, 2016
The functional pmoA gene is frequently used to probe the diversity and phylogeny of methane-oxidizing bacteria (MOB) in various environments. Here, we compared the similarities between the pmoA gene and the corresponding 16S rRNA gene sequences of 77 described species covering gamma- and alphaproteobacterial methanotrophs (type I and type II MOB ...

Erratum to: A novel pmoA lineage represented by the acidophilic methanotrophic bacterium Methylocapsa acidophila B2 [PDF]

open access: bronzeArchives of Microbiology, 2001
A fragment of the functional gene pmoA, which encodes the active-site polypeptide of particulate methane monooxygenase (pMMO), was PCR-amplified from DNA of the recently described acidophilic methanotrophic bacterium Methylocapsa acidophila B2. This methanotroph was isolated from an acidic Sphagnum peat bog and possesses a novel type III arrangement of
