Use of Pectinase in Pollen Tube and Pollen Mother Cell Smears

Styles of Physalis pruinosa L. and P. ixocarpa Brot. were fixed in a modification of Carnoy's fixative (alcohol, chloroform, acetic acid; 3:2:1) 48 hr after pollination. After 24 hr, the styles were transferred to a 5% aqueous pectinase solution (adjusted to pH 4) on a spot plate for 24 hr.

An ultrastructural study of cytomixis in tobacco pollen mother cells

Intercellular chromatin migration (cytomixis) in the pollen mother cells of two tobacco (Nicotiana tabacum L.) lines was analyzed by electron microscopy during the first meiotic prophase. The maximal manifestation of cytomixis was observed in the pachytene.

Isolation and culture studies on pollen tetrad and pollen mother-cell protoplasts

Abstract Techniques have been refined for the isolation and culture of pollen tetrad and pollen mother-cell (PMC) protoplasts from several angiosperms. The tetrads and PMC are treated with 0.75% Helicase in 10% sucrose solution at 22–25°, and within 1 h, nearly 100% of the protoplasts are released from their massive callose walls.
