Prokaryotic taxonomy in the sequencing era – the polyphasic approach revisited

Environmental Microbiology, 2011
Summary The ultimate goal of taxonomy is to establish a system that mirrors the ‘order in nature’. In prokaryote microbiology, almost all taxonomic concepts try to mirror the whole evolutionary order back to the origin of life with the cell as basic unit.

Polyphasic taxonomy ofAspergillus fumigatusand related species

Mycologia, 2005
The variability within Aspergillus fumigatus Fresenius and related species was examined using macro-, micro-morphology, growth temperature regimes and extrolite patterns. In addition, DNA analyses including partial beta-tubulin, calmodulin and actin gene sequences were used. Detailed examination of strains, considered as A.

Fluorescence Spectroscopy as a Promising Tool for a Polyphasic Approach to Pseudomonad Taxonomy

Current Microbiology, 2008
Fluorescence spectroscopy is an emerging tool for the analysis of biomolecules from complex matrices. We explored the potentialities of the method for the pseudomonad taxonomic purpose at the genus and species level. Emission spectra of three intrinsic fluorophores (namely, NADH, tryptophan, and the complex of aromatic amino acids and nucleic acid ...

Morphological studies in Rosellinia (Xylariaceae): the first step towards a polyphasic taxonomy

Mycological Research, 2005
More than 90 species are accepted in the xylariaceous genus Rosellinia. Thorough morphological characterisation of all known species will provide a taxonomic hypothesis that can be later tested by DNA sequence analysis. Exploratory analyses of ascospore morphology, shape of ascus apical structure, and anamorph genus suggest six morphological groups in ...

Polyphasic taxonomy of symbiotic rhizobia from wild leguminous plants growing in Egypt

Folia Microbiologica, 2003
About 20 strains of rhizobia from wild legumes were characterized based on numerical analysis of phenotypic characteristics, nodulating ability, fatty acid methyl esters (FAME) and SDS-PAGE profiles of whole cell proteins. FAME analysis revealed that palmitic (16:0), stearic (18:0) and arachidonic (20:0) were detected in most of wild-legume rhizobia ...

Polyphasic taxonomy and molecular identification of Australian acidovorax avenae strains

2019
The phylogenetic relationships among members of the family Comamonadaceae and several unclassified strains were studied by direct sequencing of their PCR amplified 16S rRNA genes. Based on the 16S rRNA gene sequence analysis, members of the family formed a coherent phylogenetic group.

[Study on polyphasic taxonomy of rhizobia isolated from Lespedeza species].

Wei sheng wu xue bao = Acta microbiologica Sinica, 2004
The diversity of rhizobia isolated from Lespedeza spp. was determined on the basis of numerical analysis of phenotypic characteristics, sodium dodecyl sulfate-polyacrylarmide gel electrophoresis (SDS-PAGE) of proteins, DNA-DNA homology and restriction fragment length polymorphism (RFLP) analysis of 16S-ribosomal DNA genes.

Polyphasic approach to the taxonomy of selected cyanobacteria

2015
The modern taxonomic revision of cyanobacteria is a work in progress, with both theoretical and practical challenges to be addressed. This thesis investigated selected terrestrial cyanobacterial taxa using a polyphasic approach, based on molecular phylogenetic analysis and accompanied by phenotypic characterization and nomenclatural treatment.
