Profiling Localized Immunomodulation and Drug Biodistribution within a Subcutaneous Vascularized Niche for Cell Transplantation

open access: yesAdvanced Science, EarlyView.
We studied how five common immunosuppressants behave when delivered directly to a transplant site instead of systemically. Using a vascularized implant for islet transplantation, we show that local delivery protects grafts, limits drug spread to the rest of the body, and produces distinct immune signatures.

Ammonia Detoxification Inhibits Liver Metastasis by Reshaping Hepatic Microenvironment

open access: yesAdvanced Science, EarlyView.
Liver metastasis diverts aspartate into hyperactive pyrimidine synthesis, disrupting urea cycling and causing pathogenic ammonia accumulation. Ammonia dually reprograms the microenvironment by: (1) activating hepatic stellate cells (HSCs) into pro‐fibrotic metastasis‐associated fibroblasts (MAFs), and (2) suppressing anti‐tumor monocytes/macrophages ...

Population‐Based Modeling to Predict Human PK/PD of TAK‐500, an Anti‐CCR2 Antibody–Drug Conjugate for First‐in‐Human Study in Cancer Patients

open access: yesClinical and Translational Science
TAK‐500 is a novel immune cell–directed antibody–drug conjugate (iADC) composed of TAK‐202, an anti‐CCR2 monoclonal antibody, conjugated to the STING agonist dazostinag (TAK‐676), and is designed to stimulate antitumor immunity by reprogramming CCR2 ...

Carboranyl‐Curcuminoids for the Neutron Capture‐Based Treatment of Amyloid Aggregates in Alzheimer's Disease

open access: yesAdvanced Science, EarlyView.
The 10B‐enriched monocarbonyl analog of curcumin (BMAC) 10B‐9 enables site‐specific Boron Neutron Capture Therapy (BNCT) on amyloid‐β (Aβ) fibrils. Neutron irradiation induces histidine oxidation and fibril destabilization, as revealed by 1H‐NMR and FESEM analyses.

In Vivo CRISPR Screening Identifies the Glutamate Receptor GRIA2 as Promoting Peritoneal Metastasis of Gastric Cancer via Calcium‐Dependent β‐Catenin Activation

open access: yesAdvanced Science, EarlyView.
We identified GRIA2 as a critical driver of gastric cancer peritoneal metastasis through in vivo CRISPR screening. Mechanistically, GRIA2‐mediated calcium influx inhibits GSK3β and activates Wnt/β‐catenin signaling, driven by glutamate from cancer‐associated fibroblasts.

Ligand non-competitive GITR antibody prevents formation of the obligatory signal-triggering GITRL: GITR stoichiometry

open access: yesScientific Reports
The prevalence of autoimmune diseases such as inflammatory bowel disease (IBD) and rheumatoid arthritis (RA) is increasing. Glucocorticoid-induced TNFR-related protein (GITR), a TNF receptor superfamily (TNFRSF) member, is activated by GITR-ligand (GITRL)

Leveraging Artificial Intelligence and Large Language Models for Cancer Immunotherapy

open access: yesAdvanced Science, EarlyView.
Cancer immunotherapy faces challenges in predicting treatment responses and understanding resistance mechanisms. Artificial intelligence (AI) and machine learning (ML) offer powerful solutions for cancer immunotherapy in patient stratification, biomarker discovery, treatment strategy optimization, and foundation model development.

Fully Humanized Bispecific T Cell Engager Shows Potent Activity in Central Nervous System and Peripheral Tumors

open access: yesAdvanced Science, EarlyView.
This study reports the development of a fully humanized bispecific T cell engager targeting IL13RA2, a tumor‐associated antigen enriched in glioblastoma. This off‐the‐shelf immunotherapy drives potent, antigen‐dependent T cell activation and tumor killing, and prolongs survival in experimental GBM and other solid tumors models without detectable off ...
