Preclinical Toxicity and Pharmacokinetic Evaluation of Paclitaxel Nanodispersion

AAPS PharmSciTech
This study evaluates the safety, tolerability, pharmacokinetics, and tissue distribution of paclitaxel injection concentrate for nanodispersion (PICN), either as standalone treatment or in comparison with Abraxane® (AbX) and Oncotaxel (OtX, generic formulation of Taxol® from Sun Pharma).

Topiramate: a review of preclinical, pharmacokinetic, and clinical data

Clinical Therapeutics, 1997
The antiepileptic drug (AED) topiramate is a monosaccharide derivative with a sulfamate functionality. It modulates voltage-dependent sodium conductance, potentiates gamma-aminobutyric acid-evoked currents, and blocks the kainate/AMPA (alpha-amino-3-hydroxy-5-methylisoxazole-4-propionic acid) subtype of the glutamate receptor.

Preclinical Safety and Pharmacokinetics of Recombinant Human Factor XIII [PDF]

open access: yesToxicologic Pathology, 2005
Factor XIII (FXIII) is a thrombin-activated protransglutaminase responsible for fibrin clot stabilization and longevity. Deficiency in FXIII is associated with diffuse bleeding and wound-healing disorders in humans. This report summarizes results from several studies conducted in adult cynomolgus monkeys ( M.

Exploration of the African Green Monkey as a Preclinical Pharmacokinetic Model: Intravenous Pharmacokinetic Parameters

Drug Metabolism and Disposition, 2008
The value of cynomolgus and rhesus monkeys to predict human pharmacokinetic parameters has been well established in recent years. However, practical limitations on cost and accessibility can often be a deterrent to obtain data in these valuable species, and the characterization of the predictive power of other nonhuman primates would be useful ...

Preclinical Pharmacokinetics of PDE-310, a Novel PDE4 Inhibitor

Drug Metabolism and Pharmacokinetics, 2011
A novel phosphodiesterase-4 inhibitor, 2-aryl-7(3',4'-dialkoxyphenyl)-pyrazolo[1,5-alpha] pyrimidine (PDE-310), has been synthesized for the treatment of respiratory diseases. We conducted in vitro and in vivo studies to characterize the pharmacokinetics of PDE-310.

New approaches in preclinical and clinical pharmacokinetics.

Cancer surveys, 1994
Pharmacokinetic studies are now playing an increasingly important part in the preclinical and clinical development of new anti-tumour agents. Preclinically, pharmacokinetic, metabolism and toxicological investigations are being integrated and used for the rational selection of new agents with a favourable pharmacological profile. More importantly, this

Preclinical pharmacokinetic studies of villocarine A, an active Uncaria alkaloid

Drug Testing and Analysis
AbstractVillocarine A is a bioactive indole alkaloid isolated from the Uncaria genus. It has demonstrated vasorelaxation activity and potential to protect the central nervous system. To identify the pharmacokinetic properties of villocarine A, a series of in vitro and in vivo studies have been performed.

Preclinical Pharmacokinetics: An Approach Towards Safer and Efficacious Drugs

Current Drug Metabolism, 2006
Lack of efficacy and toxicity are considered to be major reasons for drug failures and pharmacokinetics governs them to a large extent. Compound with favorable pharmacokinetics is more likely to be efficacious and safe. Therefore, the preclinical pharmacokinetic evaluation should be comprehensive enough to ensure that compounds do not fail in the ...
