Remimazolam Ameliorates Autistic‐Like Behaviors via Suppression of Ferroptosis in VTA Dopaminergic Neurons in a Mouse Model of ASD

open access: yesAdvanced Science, EarlyView.
The ultra‐short‐acting sedative remimazolam has a sustained therapeutic effect on the core symptoms of VPA‐exposed mice. Remimazolam, a GABA agonist, exerts its therapeutic effects by protecting dopamine neurons in the VTA of VPA‐exposed mice. Meanwhile, ferroptosis is the critical mechanism by which remimazolam protects VTA dopaminergic neurons and ...

Autophagy Proteins and clinical data reveal the prognosis of polycystic ovary syndrome

open access: yesBMC Pregnancy and Childbirth
Objective We aimed to investigate the significance of autophagy proteins and their association with clinical data on pregnancy loss in polycystic ovary syndrome (PCOS), while also constructing predictive models.

Tim1 Deficiency Mediates Gestational Hyperglycemia‐Related Syncytiotrophoblast Dysfunction and Fetal Growth Restriction

open access: yesAdvanced Science, EarlyView.
Hyperglycemia during pregnancy impairs the fusion of trophoblast cells into syncytiotrophoblasts, leading to fetal growth restriction. This impaired fusion is mediated by Tim1 downregulation via hyperglycemia‐induced ROS. Antioxidant therapy during pregnancy promotes syncytiotrophoblast formation by upregulating Tim1 expression, thus alleviating fetal ...

Placenta: an old organ with new functions

open access: yesFrontiers in Immunology
The transition from oviparity to viviparity and the establishment of feto-maternal communications introduced the placenta as the major anatomical site to provide nutrients, gases, and hormones to the developing fetus. The placenta has endocrine functions,

Microglia Mitochondria Support Neuronal Maturation via Metabolic and Transcriptional Reprogramming in Human 3D In Vitro Brain Model

open access: yesAdvanced Science, EarlyView.
xx xx. ABSTRACT Autism Spectrum Disorder (ASD) is a neurodevelopmental condition characterized by disrupted neuronal circuit maturation. Emerging evidence implicates microglial function and mitochondrial regulation as contributors to ASD‐associated biology, yet the mechanisms linking these processes to neuronal development remain poorly defined ...

Engineered Living Systems With Self‐Organizing Neural Networks: From Anatomy to Behavior and Gene Expression

open access: yesAdvanced Science, EarlyView.
Ectodermal tissue excised from Xenopus embryos self‐organizes into a three‐dimensional mucociliary organoid. Here, we generate a neural variant, termed neurobot, by implanting neural precursor cells. Neurobots develop mature neurons, adopt distinct morphologies, exhibit more complex motility, and respond differentially to neuroactive compounds. Imaging

On placental and lactational transfer of IgG‐based therapeutic proteins – Current understanding and knowledge gaps from a clinical pharmacology perspective

open access: yesClinical and Translational Science
Maternal medication use may expose the developing fetus through placental transfer or the infant through lactational transfer. Because pregnant and lactating individuals have been historically excluded from early drug development trials, there is often ...

Exposure to ambient fine particulate matter components during pregnancy and early childhood and its association with asthma, allergies, and sensitization in school-age children

open access: yesEnvironmental Health and Preventive Medicine
Background: Exposure to fine particulate matter (PM2.5) has been associated with allergic diseases, including asthma. However, information about the effects of specific PM2.5 components is limited.

Decoding Human Placental Cellular and Molecular Responses to Obesity and Fetal Growth

open access: yesAdvanced Science, EarlyView.
Women with obesity often deliver large‐for‐gestational‐age (LGA) infants. Single‐nucleus RNA sequencing of term placenta reveals that hypoxia and TNF‐α signaling in syncytiotrophoblasts are featured in maternal obesity, but inflammatory signatures in Hofbauer cells and response to lipid or carbohydrate metabolism in fibroblasts are specific to LGA.

Testicular mRNA‐LNP Delivery: A Novel Therapy for Genetic Spermatogenic Disorders

open access: yesAdvanced Science, EarlyView.
In the current study, we developed a novel therapy for genetic spermatogenic disorders. It was revealed that in vivo delivery of mRNA LNP3 could restore spermatogenesis in Msh5D486Y/D486Y and Maps KO mouse models with meiotic arrest. Notably, the offspring without genomic integration was born using ICSI derived from the rescue of Msh5D486Y/D486Y mouse ...
