Protective effect of stromal Dickkopf-3 in prostate cancer: opposing roles for TGFBI and ECM-1 [PDF]

open access: yes, 2018
Aberrant transforming growth factor–β (TGF-β) signaling is a hallmark of the stromal microenvironment in cancer. Dickkopf-3 (Dkk-3), shown to inhibit TGF-β signaling, is downregulated in prostate cancer and upregulated in the stroma in benign prostatic ...

Endoglin mediates the tumor‐ and metastasis‐promoting traits of stromal myofibroblasts in human breast carcinomas

open access: yesMolecular Oncology, EarlyView.
Carcinoma‐associated fibroblasts (CAFs) in tumors influence cancer progression. We identified endoglin (ENG) as a key factor in TGF‐β signaling in myofibroblastic CAFs (myCAFs), linked to poor breast cancer outcomes. Inhibiting ENG on myCAFs suppressed the TGF‐β‐Smad2/3 pathway, reducing primary tumor growth and metastasis.

Tyrol Prostate Cancer Demonstration Project : early detection, treatment, outcome, incidence and mortality [PDF]

open access: yes, 2008
This study aimed to evaluate the effectiveness of a well-controlled programme of early detection and treatment of prostate cancer in the population of Tyrol, Austria, where such a programme of early detection and treatment was initiated in 1988 and where

ITGAV and SMAD4 influence the progression and clinical outcome of pancreatic ductal adenocarcinoma

open access: yesMolecular Oncology, EarlyView.
In SMAD4‐positive pancreatic ductal adenocarcinoma (PDAC), integrin subunit alpha V (ITGAV) activates latent TGF‐β, which binds to the TGF‐β receptor and phosphorylates SMAD2/3. The activated SMAD2/3 forms a complex with SMAD4, and together they translocate to the nucleus, modulating gene expression to promote proliferation, migration, and invasion. In

The Effect of Pre-Biopsy Prostate MRI on the Congruency and Upgrading of Gleason Grade Groups Between Prostate Biopsy and Radical Prostatectomy

open access: yesSociété Internationale d’Urologie Journal
Introduction: Prostate biopsy results form the mainstay of patient care. However, there is often significant discordance between the biopsied histology and the ‘true’ histology shown on a radical prostatectomy (RP).

Is birthweight associated with total and aggressive/lethal prostate cancer risks? A systematic review and meta-analysis [PDF]

open access: yes, 2016
BACKGROUND: It has been hypothesised that intrauterine exposures are important for subsequent prostate cancer risk. Prior epidemiological studies have used birthweight as a proxy of cumulative intrauterine exposures to test this hypothesis, but results ...

Targeting carbonic anhydrase IX/XII prevents the anti‐ferroptotic effect of stromal lactic acid in prostate carcinoma

open access: yesMolecular Oncology, EarlyView.
In prostate carcinoma, lactic acid, secreted by highly glycolytic cancer‐associated fibroblasts, is imported into tumor cells through the MCT1 transporter and prevents RSL3 and erastin‐induced ferroptosis (A). Targeting of carbonic anhydrase IX/XII, the main extracellular pH regulators, in tumor and stromal cells reduces microenvironmental acidosis and

Long-term results of a phase II study of hypofractionated proton therapy for prostate cancer: moderate versus extreme hypofractionation

open access: yesRadiation Oncology, 2019
Background We performed a prospective phase II study to compare acute toxicity among five different hypofractionated schedules using proton therapy. This study was an exploratory analysis to investigate the secondary end-point of biochemical failure-free

Hyperpolarized 13C-pyruvate MRI detects real-time metabolic flux in prostate cancer metastases to bone and liver: a clinical feasibility study. [PDF]

open access: yes, 2020
BackgroundHyperpolarized (HP) 13C-pyruvate MRI is a stable-isotope molecular imaging modality that provides real-time assessment of the rate of metabolism through glycolytic pathways in human prostate cancer.

Increased expression and activity of p75NTR are crucial events in azacitidine-induced cell death in prostate cancer [PDF]

open access: yes, 2016
The high affinity nerve growth factor (NGF) NGF receptor, p75NTR, is a member of the tumor necrosis factor (TNF) receptor superfamily that shares a conserved intracellular death domain capable of inducing apoptosis and suppressing growth in prostate ...
