Ablative radiotherapy in castration‐resistant prostate cancer

open access: yesBJU International, EarlyView.
Objective To prove the oncological benefit of ablative radiotherapy in patients with up to five metastases from castration‐resistant prostate cancer (CRPC) a single‐centre randomised trial was initiated. Patients and Methods This monocentric, randomised, phase II clinical trial enrolled patients with up to five prostate‐specific membrane antigen ...

Evaluation of triplet therapy combined with robot‐assisted cytoreductive prostatectomy for high‐volume metastatic prostate cancer: a multicentre randomised controlled trial protocol

open access: yesBJU International, EarlyView.
Background Prostate cancer (PCa) is the second most common cancer in men. For patients with high‐volume metastatic PCa (mPCa) who are fit for docetaxel, a triplet therapy regimen consisting of androgen deprivation therapy (ADT), chemotherapy, and an androgen receptor pathway inhibitor (ARPI) is recommended.

Targeting KRAS for cancer therapy

open access: yesBritish Journal of Pharmacology, EarlyView.
In recent years, therapeutics targeted against KRAS proto‐oncogene GTPase (KRAS)‐mutant cancers have seen significant progress. Herein we outline the biology and epidemiology of KRAS alterations at the lineage and allele levels, reviewing the clinical evidence for KRASG12C inhibition from the discovery of the recessive switch pocket to sotorasib ...

The role of macrophage migration inhibitory factor (MIF) in the pathogenesis and progression of cardiovascular disease

open access: yesBritish Journal of Pharmacology, EarlyView.
Abstract Cardiovascular disease (CVD) remains the leading global cause of death, driven by complex mechanisms, in which chronic inflammation plays a central role. Inflammatory pathways contribute to all stages of CVD, from endothelial dysfunction and plaque formation to erosion, rupture and myocardial injury.

NRF2/xCT‐Mediated Antioxidant Adaptation Attenuates Apoptosis Induced by ASCT2 Inhibition in Prostate Cancer

open access: yesCancer Science, EarlyView.
ASCT2 inhibition induces ROS accumulation and activates NRF2‐dependent xCT upregulation in prostate cancer. This adaptive response partially supports residual cystine acquisition and glutathione synthesis, whereas combined ASCT2 and xCT inhibition disrupts redox compensation and predominantly promotes apoptotic cell death.

Hypericin Suppresses Liver Cancer Through Autophagic Degradation of AKT and Eliciting Antitumor Immune Response

open access: yesCancer Science, EarlyView.
Hypericin induces autophagic AKT degradation by promoting AKT‐HSPA5 interaction in an S473 phosphorylation‐dependent manner, suppressing HCC growth. It enhances CD8+ T cell‐mediated antitumor immunity and synergizes with anti‐PD‐L1 therapy, showing favorable safety for HCC treatment. ABSTRACT Hepatocellular carcinoma (HCC) is the most prevalent primary
