Prime editors (PEs) enhance gene editing precision but face repair mechanism limitations. DNA methylation (CpG sites) and de novo methyltransferases (DNMT3A/DNMT3B) boost PE efficiency. We developed an episomal PE system (epiPE) using EBNA1/oriP to sustain expression and methylation.

Clinical and therapeutic data of a child with ecthyma gangrenosum
The presence of the Pseudomonas aeruginosa infection in healthy children is very uncommon. Infants may occasionally present with community-acquired sepsis due to Pseudomonas aeruginosa, preceded by prolonged contact with contaminated bath water ...

Physiology of nitrate reduction in Pseudomonas aeruginosa. [Translation from: Z.allg.Mikrobiol. 13, 83-94, 1973.] [PDF]
Pseudomonas aeruginosa has for some time been known as a denitrifier. Pseudomonas aeruginosa was chosen for further studies, because P. aeruginosa occurs abundantly in Plusssee and moreover there are contradictory assertions on the gas products of ...

This study uncovers that CD38high monocytes discriminate sepsis and sterile inflammation and are associated with 28‐day mortality in bacterial sepsis. Targeting CD38 therapy reduces inflammatory response in monocytes and in sepsis mice model. Mechanistically, CD38‐HIF‐1α/glycolysis/MGO loop exacerbates sepsis‐induced immune dysregulation.

Learning interpretable models of phenotypes from whole genome sequences with the Set Covering Machine [PDF]
The increased affordability of whole genome sequencing has motivated its use for phenotypic studies. We address the problem of learning interpretable models for discrete phenotypes from whole genomes. We propose a general approach that relies on the Set Covering Machine and a k-mer representation of the genomes.

Bacteria can be genetically engineered to kill specific pathogens or inhibit their virulence. We previously developed a synthetic genetic system that allows a laboratory strain of Escherichia coli to sense and kill Pseudomonas aeruginosa in vitro.

Novel Bivalent mRNA‐LNP Vaccine for Highly Effective Protection against Pneumonic Plague
The study investigates mRNA‐lipid nanoparticle vaccines targeting Yersinia pestis, the causative agent of plague. Combined F1+LcrV mRNA vaccination elicited robust immune responses and protected mice against virulent wild‐type and atypical strains.

Trapping and Assembly of Living Colloids at Water/Water Interfaces [PDF]
We study the assembly of colloids in a two phase water-water system that provides an environment that can sustain bacteria, providing a new structure with rich potential to confine and structure living colloids. The water-water system, formed via phase separation of a casein and xanthan mixture, forms a 3-D structure of coexisting casein-rich and ...
