CRITICAL EVALUATION OF THE PSORALENS

Opinions as to the usefulness of the psoralen group of drugs in treating vitiligo were obtained by questionnaire from 100 dermatologists in various parts of the world. Two dermatologists were enthusiastic about the systemic use of the psoralens, eight were vehement in their condemnation, and most considered the results poor.

Psoralen Phototherapy and Phototoxicity

Psoralen therapy followed by exposure to long-wave ultraviolet (UV-A) light, a combination called PUVA therapy, stimulates pigmentation of the skin. PUVA therapy is effective in the treatment of vitiligo and hypopigmented scars. Because psoralens sensitize skin to UV-A light, phototoxic reactions are frequent.

Alkali reversal of psoralen cross-link for the targeted delivery of psoralen monoadduct lesion [PDF]

Psoralen intercalates into double-stranded DNA and photoreacts mainly with thymines to form monoadducts and interstrand cross-links. We used an oligonucleotide model to demonstrate a novel mechanism: the reversal of psoralen cross-links by base-catalyzed rearrangement at 90 degrees C (BCR).

Photosensitizers have been used in medicine for several thousand years. In the past, the effect being sought was repigmentation of light areas in the skin. Most of the orally administered or locally applied preparations were obtained from plants that contain psoralen (furocoumarin).

Kinetic study on pyrolysis of psoralen

In this study, products of psoralen pyrolysis were detected using a solid pyrolysis apparatus and synchrotron radiation vacuum ultraviolet photoionization mass spectrum (SVUV-PIMS). The pyrolytic kinetics of psoralen was also studied by calculating its initial pyrolytic route in quantum chemistry.
