Effect of chemotherapy on passenger mutations in metastatic colorectal cancer

open access: yesMolecular Oncology, EarlyView.
Changes in passenger mutation load and predicted immunotherapy response after chemotherapy treatment. Tumor cells rich with passenger mutations have increased sensitivity to chemotherapy. Correlation of passenger mutations with neoantigen load suggests highly mutated clones promote a more effective response to immunotherapy, and therefore, first‐line ...

Singing classes for chronic obstructive pulmonary disease: a randomized controlled trial

open access: yesBMC Pulmonary Medicine, 2012
Background There is some evidence that singing lessons may be of benefit to patients with chronic obstructive pulmonary disease (COPD). It is not clear how much of this benefit is specific to singing and how much relates to the classes being a group ...

The challenges faced in the design, conduct and analysis of surgical randomised trials [PDF]

open access: yes, 2009
Randomised evaluations of surgical interventions are rare; some interventions have been widely adopted without rigorous evaluation. Unlike other medical areas, the randomised controlled trial (RCT) design has not become the default study design for the ...

Understanding controlled trials: Randomisation methods: concealment

open access: yesBMJ, 1999
Randomisation is the best method removing selection bias between two groups of patients.1 However, the process of randomisation can be compromised such that the allocation results in biased groups of patients. A trial which has had its randomisation compromised may apparently show a treatment effect that is entirely due to biased allocation.

Methylation biomarkers can distinguish pleural mesothelioma from healthy pleura and other pleural pathologies

open access: yesMolecular Oncology, EarlyView.
We developed and validated a DNA methylation–based biomarker panel to distinguish pleural mesothelioma from other pleural conditions. Using the IMPRESS technology, we translated this panel into a clinically applicable assay. The resulting two classifier models demonstrated excellent performance, achieving high AUC values and strong diagnostic accuracy.

A Randomised Controlled Trial [PDF]

open access: yes, 2015
Liver surgery is still associated with a high rate of morbidity and mortality. We aimed to compare different haemodynamic treatments in liver surgery. In a prospective, blinded, randomised, controlled pilot trial patients undergoing liver resection were ...

Detection of circulating tumor DNA in colorectal cancer patients using a methylation‐specific droplet digital PCR multiplex

open access: yesMolecular Oncology, EarlyView.
We developed a cost‐effective methylation‐specific droplet digital PCR multiplex assay containing tissue‐conserved and tumor‐specific methylation markers. The assay can detect circulating tumor DNA with high accuracy in patients with localized and metastatic colorectal cancer.

A statistical framework for quantifying clinical equipoise for individual cases during randomized controlled surgical trials

open access: yesTrials, 2011
Background Randomised controlled trials are being increasingly used to evaluate new surgical interventions. There are a number of problematic methodological issues specific to surgical trials, the most important being identifying whether patients are ...
