Single‐molecule DNA flow‐stretch assays for high‐throughput DNA–protein interaction studies

open access: yesFEBS Open Bio, EarlyView.
We describe an optimised single‐molecule DNA flow‐stretch assay that visualises DNA–protein interactions in real time. Linear DNA fragments are tethered to a surface and stretched by buffer flow for fluorescence imaging. Using λ and φX174 DNA, this protocol enhances reproducibility and accessibility, providing a versatile approach for studying diverse ...

Analysing the significance of small conformational changes and low occupancy states in serial crystallographic data

open access: yesFEBS Open Bio, EarlyView.
This protocol paper outlines methods to establish the success of a time‐resolved serial crystallographic experiment, by means of statistical analysis of timepoint data in reciprocal space and models in real space. We show how to amplify the signal from excited states to visualise structural changes in successful experiments.

Sudden anaerobization in Amphibacillus xylanus increases intracellular labile ferrous iron and inhibits cell growth

open access: yesFEBS Open Bio, EarlyView.
Abruptly changing from aerobic to anaerobic conditions (sudden anaerobization) induced growth inhibition and a significant increase in intracellular labile ferrous iron in the aerotolerant anaerobe Amphibacillus xylanus. We found that free flavins mediate efficient electron transfer from NADH to ferric iron under anaerobic conditions, suggesting that ...

RoundMi: A quantitative method to analyze mitochondrial morphology in mitotic cells

open access: yesFEBS Open Bio, EarlyView.
RoundMi is a workflow for rapid analysis of mitochondrial morphology in mitotic cells. By combining adaptive preprocessing with automated segmentation and quantification, it enables accurate measurements from single focal plane images, reducing acquisition time and computational demands while remaining compatible with high‐throughput fixed and live ...

Development and testing of a high-precision static calibration device for aviation thin-film thermocouples

open access: yesScientific Reports
This study aims to develop a novel calibration device to address the challenges of inadequate temperature field uniformity, significant temperature fluctuation, and less reliability in existing static calibration devices for thin-film thermocouples.

Versatile vector tools for efficient protein screening across multiple expression systems

open access: yesFEBS Open Bio, EarlyView.
A unified vector toolkit enables rapid protein expression screening across E. coli, insect, and mammalian cells. A single primer pair amplifies the target gene, which is inserted into any vector via a standardized interface. This streamlined workflow eliminates repeated cloning steps, accelerating the identification of optimal expression conditions for

Calculation of a Reference Interval for Rectal Temperature in Adult Dogs Presenting for Veterinary Care Using an Algorithm for Mixed Data

open access: yesAnimals
Veterinarians rely on the measurement of canine body temperature to define the health status of dogs, but no studies exist defining a reference range for rectal temperature on a large group of dogs.

Time‐restricted feeding prior to Mycobacterium tuberculosis infection reduces tissue CD4+ T cells with limited impact on bacterial clearance

open access: yesFEBS Open Bio, EarlyView.
Time‐restricted feeding (TRF) in mice increased liver fatty acid oxidation and decreased fatty acid biosynthesis. These alterations persisted when TRF was discontinued and the host was infected with Mycobacterium tuberculosis. Pre‐exposure to TRF did not alter tissue (lung and spleen) mycobacterial burden but significantly reduced CD3+ T cells in lungs

A bandgap reference source with higher-order curvature compensation

open access: yesDianzi Jishu Yingyong
Based on the 0.18 μm BCD process, a higher-order curvature compensation bandgap reference with low temperature coefficient is proposed. On the basis of the basic Kuijk bandgap reference structure, the VBE linearization method is used to perform high ...
