Advanced Science, EarlyView.An enzyme–nitric oxide (NO) co‐regulatory strategy reprograms pathological extracellular matrix (ECM)–fibroblast crosstalk in hypertrophic scars. Enzymatic degradation of pre‐existing ECM relieves aberrant mechanical cues, while NO‐mediated fibroblast phenotypic modulation restrains excessive collagen synthesis.
