A series of 2‐substituted adenosine 5′‐O‐triphosphate derivatives is synthesized and tested as substrates for polymerases. In vitro transcription with T7 RNA polymerase, primer extension with engineered DNA polymerases, or 3′‐labelling with poly(A) polymerase are used for enzymatic synthesis of base‐modified RNA.
