An Engineered PfAgo with Wide Catalytic Temperature Range and Substrate Spectrum
This work presents a strategy for engineering thermophilic Ago proteins to obtain wide catalytic temperature range and broad substrate spectrum. This strategy is applied to engineer PfAgo and TtdAgo, and the biochemical properties of the engineered PfAgo are systematically characterized. The study further elucidates the structural differences involved.

Collaborative Duality of CircGLIS3(2) RNA and Protein in human Wound Repair
CircGLIS3(2), a circRNA transiently upregulated in dermal fibroblasts during acute wound healing, is induced by IL‐1α and TGF‐β. A new amphipathic molecule, S416 is discovered, that locks the SARS‐CoV‐2 spike protein in its closed state, blocking viral entry. S416 acts as a molecular bolt, binding six sites: three between adjacent RBDs and three connecting NTDs to RBDs. This dual‐locking mechanism stiffens the spike structure and reduces its flexibility.
