Synthetic Strategy for mRNA Encapsulation and Gene Delivery with Nanoscale Metal‐Organic Frameworks

open access: yesAdvanced Functional Materials, EarlyView.
This research utilizes the ZIF‐8 for the encapsulation and intracellular delivery of nucleic acids, specifically mRNA, for applications in gene delivery. Integrating PEI addresses the issue of mRNA leakage from ZIF‐8, resulting in the delivery and expression of green fluorescent protein (GFP) in vitro and firefly luciferase in vivo.

Functional plasticity of antibacterial EndoU toxins. [PDF]

open access: yes, 2018
Bacteria use several different secretion systems to deliver toxic EndoU ribonucleases into neighboring cells. Here, we present the first structure of a prokaryotic EndoU toxin in complex with its cognate immunity protein.

Endocytic Programming via Porous Silicon Nanoparticles Enhances TLR4 Nanoagonist Potency for Macrophage‐Mediated Immunotherapy

open access: yesAdvanced Functional Materials, EarlyView.
Porous silicon nanoparticles (PSiNPs) reprogram macrophage endocytosis of manganese@albumin‐based TLR4 nanoagonists, driving TRIF‐biased TLR4 signaling, eliciting robust proinflammatory responses, and potentiating macrophage‐mediated immunotherapeutic effects against NSCLC.

Neonatal Diet Impacts Circulatory miRNA Profile in a Porcine Model

open access: yesFrontiers in Immunology, 2020
microRNAs (miRNAs) are conserved non-coding small nucleotide molecules found in nearly all species and breastmilk. miRNAs present in breastmilk are very stable to freeze-thaw, RNase treatment, and low pH as they are protected inside exosomes.

Hamster leukemia virus: lack of endogenous DNA synthesis and unique structure of its DNA polymerase [PDF]

open access: yes, 1974
Infectious hamster leukemia virus (HaLV) contains a DNA polymerase different from those of murine and avian viruses. No endogenous reaction directed by the 60 to 70S RNA of HaLV could be demonstrated in detergenttreated HaLV virions, nor could the ...

Cholesterol in mRNA‐Lipid Nanoparticles can be Replaced with the Synthetic Mycobacterial Monomycoloyl Glycerol Analogue MMG‐1

open access: yesAdvanced Functional Materials, EarlyView.
This study demonstrates that cholesterol in messenger RNA‐lipid nanoparticles (mRNA‐LNPs) can be completely replaced with an immunopotentiating lipid, i.e., a synthetic analogue of the C‐type lectin receptor agonist monomycoloyl glycerol (MMG‐1), without compromising physicochemical properties, in vivo transfection efficiency, and immunogenicity of the

Implementation of Accurate and Fast DNA Cytometry by Confocal Microscopy in 3D

open access: yesCellular Oncology, 2005
Background: DNA cytometry is a powerful method for measuring genomic instability. Standard approaches that measure DNA content of isolated cells may induce selection bias and do not allow interpretation of genomic instability in the context of the tissue.

Quantifying RNA and DNA in planktonic organisms with SYBR Green II and nucleases. Part A. Optimisation of the assay

open access: yesScientia Marina, 2005
Assay protocols for RNA and DNA in crude plankton extracts using the fluorochrome SYBR Green II are developed here. The method is based on the fluorescence in 3 aliquots: the first measures RNA after DNA digestion; the second measures DNA after RNA ...
