Isothiazologuanosine (tzG) as Environmentally Sensitive Fluorescent Reporter in RNA

The emissive isothiazologuanosine analog tzG is synthesized for the first time as RNA phosphoramidite building block for solid‐phase synthesis. Due to its isomorphic and isofunctional characteristics, tzG forms Watson–Crick base pairs and can be used as fluorescent probe to monitor (deoxy)ribozyme activity.

Differential Water Networks Guide Selectivity Optimization of a Cell Active BPTF Inhibitor in Neuroblastoma

BZ2 is a second‐generation Bromodomain PHD finger Transcription Factor (BPTF) inhibitor with improved selectivity over Class I and Class IV BRD off‐targets identified in our previously reported inhibitor, BZ1. Structural analyses reveal that water network engagement is key to this selectivity.

Remote Desaturation of Unprotected Amines and C─H Amination of Alkanes by Polarity‐Matched N‐Radical Cations

Highly electrophilic N‐radical cations enable efficient and selective H‐atom transfer (HAT) of unprotected amines. In this exceedingly practical strategy, simple CuCl catalyzes an interrupted Hofmann–Löffler–Freytag (HLF) reaction to afford δ desaturated amines, aza‐heterocycles, and C─H aminated alkanes without protecting groups.

Quantifying Structure‐Dependent Rigidity of 3D DNA Origami Boxes

We investigate the mechanical stability of a hollow 3D DNA origami box during assembly using liquid AFM imaging and finite element simulations. As structural completeness increases from flat sheets through intermediate assemblies to a closed box, rigidity improves.
